The effects of Zn2+ on GABAA-receptor mediated responses in acutely isolated rat sacral dorsal commissural nucleus (SDCN) were studied using nystatin-perforated whole cell rechniques .The results demonstrated that (1) GABA induced inward currents through activation of GABAA~at a holding potential of-40 mV; (2) GABAA-receptor mediated responses were suppressed by Zn2+ in a reversible and voltage-independent manner; and (3) in the presence of Zn2+, the concentration-response curve of CABA-induced responses was shifted to the right in a parallel manner. The results suggest that Zn2+allosterically depresses GABAA-receptor mediated currents.

目的 明确人鼻粘膜上皮细胞Na+通道的特性 ,为研究Na+通道在鼻粘膜病理性改变及治疗中的作用奠定理论基础。方法 利用膜片钳技术对经无血清气 液界面培养的鼻源性鼾症患者手术切除下鼻甲标本的鼻上皮细胞进行Na+通道基本特性研究。结果 在细胞贴附式膜片上 ,可记录到典型的单通道电流 ,其电导为 2 1 0 9pS ,反转电位为 - 50 96mV ,且 77 78%反转电位 <- 4 0mV ,离子通透性比率PNa/PK>5 80。在Na+通道抑制剂 10 4 mmol/LAmiloride存在于电极液内时 ,Na+通道发生率从 2 6 7%减少到 5 13% (P <0 0 5) ,表明大部分Na+通道可被Amiloride抑制。 10 3mmol/LCa2 +存在于浴液时 ,Na+通道的发生率无明显增加 (P >0 0 5)。电压对开放概率无明显影响。结论 在细胞贴附式膜片上 ,人鼻粘膜上皮细胞具有大部分Amiloride敏感型的Na+选择性通道及小部分Amiloride非敏感型的Na+通道。细胞外液的Ca2 +不能激活Na+选择性通道的开放 ,通道的开放概率是非电压依赖性的

AIM To investigate the relationship between protein tyrosine phosphorylation and volume-regulated chloride channel currents in A10 vascular cells. METHODS Whole cell patch-clamp recording technique was used. RESULTS ①The change of the solution from isotonic to hypotonic caused the activation of a large, outward rectifying current based on the basal current, which exibited a slight time-dependent inactivation at positive potentials (≥+60 mV). The reversal potential of the current was( -1.5±3.4) mV, consistent with the equilibrium potential of Cl - (E Cl=0 mV). The decrease in o made the reversal potentials shift to positive potentials. The hypertonic solution inhibited the current. ②The chloride channel blocker 4,4′-diisothiocyanato-stilbene-2,2′-disulphonic acid(DIDS, 100 μmol·L -1) reversibly inhibited the current in voltage-dependent manner. At the membrane potentials of +80 mV and -80 mV, the inhibitory rates were 53.7%±6.2% and 29.8%±4.9% respectively. ③Protein tyrosine kinase inhibitor genistein (30 μmol·L -1) decreased the current in voltage-independent manner. At the membrane potentials of +80 mV and -80 mV, the currents were decreased by 62.3%±11.3% and 66.9%±10.5% respectively. ④Protein tyrosine phosphatase inhibitor sodium orthovanadate (Na 3VO 4, 500 μmol·L -1) enhanced the current activated by hypotonic solution in voltage-independent manner. It increased the current by 44.8%±14.5% and 47.1%±13.6% at the membrane potentials of +80 mV and -80 mV respectively. CONCLUSION Volume-regulated chloride channels exist in A10 cell line. Protein tyrosine phosphorylation modulates the activation of volume-regulated chloride current in A10 cells.
