Objective To establish a chronic glaucoma model in Wistar rats. Methods Chronic glaucoma model was created in 21 Wistar rats by cauterizing episcleral vessels. Three episcleral vessels of the left eye of each rat were cauterized, and the right eye was used as contol. The intraocular pressure (IOP) in both model and control eyes was measured and recorded regularly. Eyes of experimental rat were enucleated in 2 ,4 and 6 months after operation to observe the changes of optic disk and retina,and numbers of retinal ganglion cells in central and peripheral retina were counted. Results The average IOP was (35. 0±1. 6) mmHg in model eyes with an approximately 1. 8-fold more than that of control eyes (19. 4±1. 1) mmHg, and elevated IOP persisted for at least 6 months after operation. No any complication in the anterior segment was seen in model eyes. Retinal thickness became thinner with an about 46% loss of retinal ganglion cells in the peripheral retina,and 36% in the central retina. C/D of model eye was over 0. 7 after 6 months. Conclusion Establishment of chronic glaucoma model by unilaterally cauterizing the episcleral vessels in Wistar rats offers a reproducible and quantitative way for glaucomatous pharmacology study, observation of damage of optic nerve and neuroprotection as well as treatment of the optic neuropathy associated with glaucoma.

目的 利用大鼠建立慢性青光眼模型。方法 选用成年Wistar大鼠,左眼为实验眼,右眼为对照眼,找到实验眼的4条表层巩膜静脉,烧灼其中3条。定期测量和记录双眼眼压,观察眼前节情况;测量和比较双眼角膜水平直径大小。分别于烧灼后2、4、6个月时摘除双侧眼球。观察大鼠视网膜及视盘的组织形态学改变、双眼视网膜厚度和视网膜神经节细胞数目变化。结果 (1)一次性实验操作可使眼压持续升高至少6个月。(2)眼前节无明显并发症。(3)实验期间,角膜水平直径大小无改变。(4)视网膜厚度明显变薄,视网膜神经节细胞数明显减少,视盘凹陷逐渐扩大和加深。 结论 通过烧灼大鼠表层巩膜静脉可获得长期眼压中度升高的慢性青光眼模型。

Tono-pen XL Tonometer was used to measure the intraocular pressure(lOP) to gurantee IOP value at 3 day, 1 week, 1,2 3, 6 month in both model and control eyes. Eyes of experimental rat were enucleated in 1, 3 and 6 months after operation to observe the changes of retina, optic nerve and numbers of retina ganglion cells of central and peripheral retina to judge whether the model was successful. Results:After the laser treatment,42 rats model with intraocular hypertension were obtained. The angle of anterior chamber were narrow, and the fibroblasts aggregation in trabecular reticulum, the iris were also abnormal obviously. Retina thickness became thinner with obviously loss of retina ganglion cells in the central and peripheral retina, Conclusion:Establishment of chronic glaucoma model by unilaterally photocoagulating the trabecular meshwork and episcleral veins in SD rats offers a reproducible and quantitative way for glaucomatous study. Compared with the control eyes, the model eyes present characteristic glaucomatous morphological changes.

目的:建立532—二极管激光光凝的大鼠慢性高眼压性青光眼模型,评价模型眼的相关组织病理学改变。方法:选用成年SD大鼠50只,采用532-二极管激光行角膜缘小梁网和角膜缘颞侧及颞上、颞下3条巩膜浅层静脉光凝。定期测量和记录双眼眼压,观察眼前节情况。于术后1、3,6个月时摘取双侧眼球,观察双眼视网膜组织形态学改变、视网膜厚度及视网膜神经节细胞数目的改变。结果:(1)大鼠眼内压于1个月时达到眼压峰值,高眼压状态持续2月余,然后眼压水平逐渐下降,6个月时眼压基本恢复到正常范围;(2)视网膜厚度明显变薄,视网膜神经节细胞数明显减少。结论:532-二极管激光光凝SD大鼠小梁网及巩膜浅层静脉血管可以建立长期眼压中度升高的慢性高眼压性青光眼模型,模型眼出现了青光眼眼底特征性的形态学改变。

ObjectiveTo establish a simple,economic and practical chronic experimental glaucoma model has an important significance in the basic study and clinical research of glaucoma.The purpose of this paper was to creat a chronic glaucoma model by laser irradiating trabecular meshwork(TM) via external limbus and assess the damage of retinal ganglion cells.MethodsKrypton laser was used to target at the TM in the right eyes of 20 Wistar rats with the wavelength 532nm,light spot diameter 200μm ,duration 0.7s and power 0.4W.The left eyes without treatment were as control group. 80-100 spots were set along the 360° of eyeball for 2 times at a 7-day interval.Intraocular pressure(IOP) was measured using a tonometer at different times following photocoagulation of laser.The average density of retinal ganglion cells(RGCs) in photocoagulated eyes and control eyes was compared.ResultsIOP in the photocoagulated eyes were (25.30±9.40),(27.15±6.02),(26.12±8.63),(26.20±2.69),(26.15±3.29) and (25.55±1.20)mmHg at 3 days after the first laser and at 3,7,14,28,35 days after the second laser respectively.IOP in the control eyes were (12.25±3.54),(12.30±3.48),(12.28±3.76),(12.14±3.53),(12.36±3.72) and (12.60±3.49) mmHg,showing that IOP of experimental eyes at various times after laser were significantly higher than that before laser(P<0.01).The average density of RGCs in the photocoagulated eyes was (1992±108) cells/mm 2 ,and that of the control eyes was (2516±196) cells/mm 2 ,indicating a significantly lower density value in the photocoagulated eyes in comparison with control eyes at 35 days after the second irradiation(P<0.01).ConclusionPhotocoagulation of TM with translimbal laser to elevate the IOP is a simple and an effective method of establishing chronic experimental glaucoma model.

目的建立一种慢性青光眼模型,为青光眼的基础和临床科研提供实验条件。方法使用波长532nm的氪离子黄光通过角膜缘光凝20只Wistar大鼠的右眼小梁网,左眼为对照眼。分别于光凝前,光凝后不同时间测量眼压,检测并比较视网膜神经节细胞(RGCs)的丢失程度。结果第1次激光光凝后3d,第2次光凝后3、7、14、28、35d,实验眼光凝后不同时间眼压较光凝前明显升高(P<0.01)。第2次光凝后35d光凝眼视网膜铺片RGCs平均密度为(1992±108)个/mm2,对照眼RGCs平均密度为(2516±196)个/mm2,光凝眼较对照眼明显减少(P<0.01)。结论激光光凝损伤小梁网引起眼压升高是建立慢性青光眼模型的一种简单有效的方法。
