Immunohistochemical staining method(ABC method) is used to detect the Bcl 2,P53,P21 and Rb gene protein in 40 cases of primary brain gliomas and 5 cases brain tissue.Results showed:the positive rates of Bcl 2 protein,P53 protein expression were remarkably different in I to Ⅳ stage of brain gliomas.The expression of P21 protein,Rb protein was gradually increased from Ⅰ to Ⅳ stage of brain gliomas.Therefore our data indicated the Bcl 2,P53,P21 and Rb gene were important in arising of gliomas,particularly Bcl 2,P53 gene.The expression of these gene were correlated with the degree of malignant of gliomas.Detection of expression of Bcl 2,P53,P21 and Rb gene may be a useful tool for diagnosis,distinguish diagnosis and asserted of malignant and prognosis.

采用原癌基因Bcl-2、P21及抑癌基因P53、Rb蛋白4种抗体,用ABC免疫组织化学方法对40例脑星形胶质瘤组织作免疫组化染色.结果发现:Bcl-2及P53基因表达在Ⅰ~Ⅳ级胶质瘤中阳性率有显著性差异,而且染色强度也有明显差异.P21、Rb基因表达在Ⅰ~Ⅳ级胶质瘤中呈上升趋势,但仅在Ⅰ~Ⅲ级、Ⅰ~Ⅳ级之间有明显差异.可以认为:Bcl-2、P53、P21、Rb在胶质瘤的发生中发挥一定的作用,尤其是Bcl-2、P53基因,这些基因蛋白的表达与胶质瘤恶性程度有关.测定胶质瘤中的Bcl-2、P53、P21、Rb蛋白对于胶质瘤的诊断、鉴别诊断以及判断肿瘤的恶性程度与预后均有一定意义.

目的探讨与细胞周期调节相关的基因p06和Rb基因在贲门癌中的表达及其意义。方法用SP免疫级化法对58例贲门癌进行p16和Rb基因蛋白的检测。结果随着贲门癌恶性程度的加重和浸润的加深,p16基因蛋白的阳性表达率逐渐降低,而Rb基因蛋白的阳性表达率逐渐升高(P<0.05)。p16和Rb基因蛋白的表达呈显著负相关(P<0.05)。结论p16和Rb基因蛋白的表达异常在贲门癌的发生发展中起重要作用。p16基因蛋白对Rb基因蛋白的表达具有负性调节作用。

Purpose To study the expression of p16 and Rb gene proteins in primary lung cancers. Method Eightynine cases of lung cancer were observed by immunohistochemistry. Results The loss of p16 protein in lung cancers was related to histological type, lymph node metastasis and progressive stage, while the loss of Rb protein only to histological type. Purpose To study the expression of p16 and Rb gene proteins in primary lung cancers. Method Eightynine cases of lung cancer were observed by immunohistochemistry. Results The loss of p16 protein in lung cancers was related to histological type, lymph node metastasis and progressive stage, while the loss of Rb protein only to histological type. The inverse correlation of p16 and Rb gene expression in lung cancers was confirmed. Conclusion The loss of p16 or Rb gene protein is an important molecular event in lung cancers, and closely related to the block in the negative regulatory pathway of cell cycle.

目的:研究p16和Rb基因蛋白的表达与肺癌临床病理学特征的关系。方法:采用免疫组织化学方法对89例肺癌进行了p16和Rb蛋白的定位观察。结果:肺癌p16基因蛋白的总丢失率为472%(42/89),且与肺癌的组织学类型、淋巴结转移、临床病理分期有关。Rb基因蛋白的总丢失率为315%(28/89),与组织学类型有关。p16和Rb基因蛋白的表达存在着明显的互补性。结论:p16和Rb基因蛋白的丢失是肺癌发生发展中重要的分子事件,与细胞周期G1→S期的负反馈调节环路中断关系密切。
