    本研究采用系膜细胞培养技术观察了38例IgA肾病患者血清对系膜细胞增殖的影响,并采用ELlSA方法检测了血清中循环免疫复合物(CIC)的浓度,结果发现,IgA肾病血清对系膜细胞的增殖有明显刺激作用,而且这种刺激作用与肾组织病理改变程度成正相关,IgA肾病血清中IgA-CIC和IgG-CIC均较正常人明显增高,但血中IgA水平与系膜增殖及病理改变程度无相关性。

    The expression of proliferating cell nuclear antigen (PCNA)/cyclin is correlated with the S-phase of cell cycle and plays a critical role in the initiation of cell proliferation. The quantitative analysis of the expression of PCNA/cyclin in cultured rat mesangial cell was observed by immunohistochemical method in this study. It was found that Emodin had an inhibitory effect on the expression of PCNA/cyclin in mesangial cells, the number of S-phase cells was dramatically decreased when Emodin was used. Our previous work has demonstrated that Emodin can inhibite the expression of c-myc mRNA and the proliferation of mesangial cells. These results are consistent with the idea that inhibitory expression of c-myc mRNA and the effect of its genetic production is a central mechanism of Emodin leading to the inhibition of mesangial cell proliferation.

    本文应用体外细胞培养和免疫组化技术,以增殖细胞核抗原(PCNA/cyclin)为标志抗原,观察了大黄素对肾小球系膜细胞周期调控的影响,发现大黄素能明显抑制系膜细胞向S期的转化。我们以往的工作已证实大黄素能抑制肾小球系膜细胞c-myc原癌基因mRNA的表达,并可能通过阻抑系膜细胞由G_1期向S期转化,进而抑制系膜细胞增殖。本项研究通过观察大黄素对细胞周期S期标志抗原(PCNA/cyclin)的影响,进一步证实了上述理论。
