Objective To investigate the effects of Arg Gly Asp Ser (RGDS) tetrapeptide on the expressions of apoptosis related genes interleukin 1 β converting enzyme (ICE) and bcl 2 in human glomerular mesangial cells, so as to provide experimental evidence for regulating proliferation and apoptosis of mesangial cells with RGD containing polypeptide.Methods Propidium iodide staining was used to observe the condensation and fragmentation of apoptotic nuclei. The internucleosomal DNA fragmentation was detected by DNA electrophoresis on agarose gel. The expressions of ICE and Bcl 2 were determined by reverse transcription polymerase chain reaction (RT PCR).Results After ten hours of incubation, the glomerular mesangial cells treated with RGDS peptide displayed features consistent with apoptosis, such as nuclear condensation and fragmention, and a regularly spaced "ladder" shape of genomic DNA electrophoresed. And RT PCR showed that expressions of both ICE and Bcl 2 mRNAs were apparently increased in RGDS treated group, compared with that of the control group.Conclusion These results demonstrated for the first time that RGDS peptide was able to induce apoptosis in human glomerular mesangial cells, during the process ICE and bcl 2 genes may have an important regulating role.

目的探讨精氨酸-甘氨酸-天门冬氨酸-丝氨酸(RGDS)四肽对肾小球系膜细胞凋亡相关基因白细胞介素-1β-转化酶(ICE)和bcl-2表达的影响,为利用RGDS四肽调控系膜细胞的增殖与凋亡提供实验理论依据。方法合成RGDS四肽,采用经典的肾小球系膜细胞培养方法进行体外研究;碘化丙啶染色法观察凋亡细胞细胞核的固缩、碎裂现象;琼脂糖凝胶电泳法观察凋亡细胞核小体DNA的断裂现象;逆转录-聚合酶链反应技术检测凋亡相关基因ICE和bcl-2的表达变化。结果与RGDS四肽孵育10小时后,肾小球系膜细胞出现了典型的凋亡征象,诸如细胞核固缩、碎裂和DNA梯形条带等;同时,RGDS四肽处理组的ICE和bcl-2表达亦明显高于对照组。结论在RGDS四肽诱导的肾小球系膜细胞凋亡过程中,ICE和bcl-2基因对细胞凋亡可能具有重要的调节作用。

The 3H TdR incorporation of the AngⅡ plus benazeprilat group (free FCS medium plus Ang Ⅱ and benazeprilat) was lower than that of the AngⅡ group; 3. The stimulative effect of Ang Ⅱ and the inhibitory effect of benazeprilat of proliferation in glomerular mesangial cells of SHR were both dose dependent.\ Conclusion\ Benazeprilat may play an inhibitory role in the proliferation induced by Ang Ⅱ in glomerular mesangial cells of SHR. Benazeprilat has a renal protective effect at cellular level.

目的观察转换酶抑制剂(ACEI)苯那普利拉(Benazeprilat)对自发性高血压大鼠(SHR)肾小球系膜细胞增殖的影响.方法采用细胞培养和3H-胸腺嘧啶核苷(3H-TdR)参入技术.结果(1)血管紧张素Ⅱ(AngⅡ)组的3H-TdR参入率高于对照组;(2)AngⅡ加Benazeprilat组的3H-TdR参入率低于AngⅡ组;(3)AngⅡ对SHR肾小球系膜细胞增殖的刺激作用和Benazeprilat的抑制作用呈剂量依赖关系.结论苯那普利拉能逆转AngⅡ对SHR肾小球系膜细胞的增殖作用,从细胞水平体现其肾脏保护作用.
