Abstract Expressions of Rb and p53 genes in early and late 2BS cells were analyzed. 我们以不同代龄的人胚肺二倍体成纤维细胞(简称2BS细胞)为实验对象,通过斑点杂交和Northern杂交方法,观察了年轻及衰老细胞中Rb、p53基因的mRNA转录水平以及表皮生长因子(EGF)刺激对Rb、p53基因转录的影响。结果表明,衰老细胞中Rb基因的mRNA表达水平与年轻细胞基本相同,但衰老细胞中p53基因的转录水平较年轻细胞低。EGF刺激后,Rb表达水平增加,年轻细胞为305%,衰老细胞则为52%,远没有年轻细胞敏感;EGF对2BS细胞中p53基因的表达无影响。上述结果从生长因子诱导角度为探讨衰老机制提供了新的线索。

本文以大肠正常粘膜为对照,利用Northern印迹杂交、斑点杂交技术,检测了大肠癌、腺瘤中肿瘤抑制基因(RB和p53)的表达情况。实验结果表明:大多数大肠癌有RB基因、p53基因或二者同时存在的过表达现象,在个别腺瘤中也可见到这种现象。推测:可能由于RB、p53基因发生了点突变,导致基因产物失活,引起细胞增殖活动增强,反过来通过某种调节机制诱导RB基因出现过表达现象。

应用限制性内切酶片段长度多态性分析了32例原发急性淋巴细胞白血病Rb基因的等位基因变化,采用32P-标记的RbcDNA3.8kb探针,结果在8例患者中发现Rb基因结构异常(25%).其中4例出现新的3.1kb和2.3kb片段,选用Rb基因外显子18,19,21,22,27五对引物,对已发现Rb基因异常的6例患者进行了分析,发现1例在外显子18和21无扩增产物.结果提示:Rb基因异常与急性淋巴细胞白血病的疾病加剧的关系较疾病的始发关系更为密切,值得进一步研究
