The deletion and rearrangement of p53 and Rb genes were studied in gastric cancer with Southern blot hybridization and the point mutation in 248 and 249 codons of p53 was studied with PCR-RFLP.It was found that deletion or rearrangement of p53 was seen in 9 cases out of 30 cases of gastric cancer and point mutation at 248 codon of p53 in 2 cases out of 42 cases,and deletion and rearrangement of Rb gene were seen in 2 cases out of 15 cases.

本文使用Southern杂交方法分析了胃癌组织中p53基因和Rb基因的缺失和重排;使用PCR-RFLP技术分析了p53基因第248位、249位密码子的点突变。研究结果显示:30例胃癌组织中9例有p53基因的缺失或重排;42例胃癌组织中两例有p53基因第248位密码子的点突变;15例胃癌组织中两例有Rb基因的缺失和重排。

Rb gene is a cancer suppressor gene. Its product, Rb protein has close relation with the regulation of cell transformation, cell cycle and transcription. A lot of carcinomas such as retionblastoma, brest cancer, small cell lung cancer, prostate cancer, bladder cancer, oesteosacoma, soft tissue sarcoma etc. are related to the deletion or inactivation of Rb gene. Rb gene is a cancer suppressor gene. Its product, Rb protein has close relation with the regulation of cell transformation, cell cycle and transcription. A lot of carcinomas such as retionblastoma, brest cancer, small cell lung cancer, prostate cancer, bladder cancer, oesteosacoma, soft tissue sarcoma etc. are related to the deletion or inactivation of Rb gene. Stomach cancer is a very common carcinoma of the gastrointestinal system. Researchers have shown that the development of stomach cancer is due to the activation of many oncogenes and the inactivation of antioncogenes. By Southern Blot hybridization, we have tested cancer tissues of nine stomach cancer patients and one cell line BGC-823 with Rb gene probe, finding that the Rb gene of all the tissues and the cell line are abnormal as compared with that of normal human blood tissues. Among the cancer tissues of the nine patients, five have partial deletion of Rb gene, three as well as the cell line have complete deletion of Rb gene. This indicates that the loss of Rb gene is the key link of cancer development. We believe that, with the more sensitive method of Polymerase Chain Reaction (PCR), we can test the fallen cells of stomach hoping that it would be helpful to the early diagnosis of stomach cancer. Meanwhile, this experiment provides target cells of gene therapy research of tumors.

Rb基因是一种抗癌基因,其产物Rb蛋白与细胞转化、细胞周期调节及转录调控有密切关系。许多肿瘤如视网膜母细胞瘤、乳腺癌、小细胞肺癌、前列腺癌、膀胱癌、骨肉瘤、软组织肉瘤等,都与Rb基因的失活或缺失有关。胃癌是消化道常见多发肿瘤,研究表明,其发生发展与多种癌基因的活化、抗癌基因的异常有关。我们用Southern印迹杂交法,以Rb基因为探针,检测了九例胃癌患者的癌组织和一个胃癌细胞株,发现它们与正常人血相比Rb基因全部呈现异常。其中5例发生Rb基因部分缺失,3例及人胃腺癌细胞株BGC-823全部缺失。这表明在胃癌发生过程中Rb基因的丢失是一个重要环节。我们相信用更灵敏的多聚酶链式反应(PCR)技术检测胃脱落细胞,对患癌危险性的早期预报会有很大帮助。同对,本实验也对肿瘤的基因治疗研究提供了靶细胞。

Abstract Expressions of Rb and p53 genes in early and late 2BS cells were analyzed. The expression of Rb gene was relatively constant in early and late 2BS cells by Northern analysis of cellular mRNA, whereas that of p53 gene in old cells decreased compared with that in young cells by total RNA dot-blot and Northern blot.After both cells were treated by EGF, Rb RNA levels increased 305% in young cells and 52% in old ones,however, no significant change was found in p53 gene...

This research may provide a new clue to understanding the molecular mechanism of ageing.

我们以不同代龄的人胚肺二倍体成纤维细胞(简称2BS细胞)为实验对象,通过斑点杂交和Northern杂交方法,观察了年轻及衰老细胞中Rb、p53基因的mRNA转录水平以及表皮生长因子(EGF)刺激对Rb、p53基因转录的影响。结果表明,衰老细胞中Rb基因的mRNA表达水平与年轻细胞基本相同,但衰老细胞中p53基因的转录水平较年轻细胞低。EGF刺激后,Rb表达水平增加,年轻细胞为305%,衰老细胞则为52%,远没有年轻细胞敏感;EGF对2BS细胞中p53基因的表达无影响。上述结果从生长因子诱导角度为探讨衰老机制提供了新的线索。
