Objective: To study the correlation between abnormality of Rb gene in initiation of oral squamous cell carcinoma (SCC), and to analyze the relationship between deletion of Rb and some clinicopathological parameters.Method: DIGHighPrime Labeled Rb 36Kb DNA probe in combination with antiDIGAP detection system was used in this study.Results: The structural abnormal alterations of Rb gene were found in 7 cases of total samples, in which observed were the deletion of 54, 41 and 23Kb fragment in 6 cases. Rearrangement was detected in one case. Further analyses were carried out to determine whether the deletion of Rb gene correlated with various clinicopathological parameters of the patients. No correlation was found with sex, age, site of primary tumor, level of histopathological differentiation and TNM staging.Conclusion: The abnormalities of Rb gene might be associated with the carcinogenesis of oral SCC.

Objective To study the significance of c-myc oncogene and retinoblastoma (Rb) tumor-suppressor gene in the tumorigenesis of oral squamous cell carcinoma(SCC) and in the malignant changes of oral premalignant lesions.Methods c-myc oncogene protein and Rb gene protein expression in 40cases of oral SCC and 10 cases of leukoplakia as well as 14 cases of lichen planus were quantitatively studied by flow cytometry and cellular immunofluorescence staining technique.Results The c-myc oncogene expression was found in 65%,30% and 21% of the oral SCC,leukoplakia and lichen planus,respectively. The obvious different statistical significances of the fluorescence indexes(FI) in c-myc oncogene expression were found between the oral SCC and leukoplakia as well as lichen planus.(P< 0.05 and P< 0.005 ).The Rb gene protein was detected in 30%,0 and 7% of the oral SCC,leukoplakia and lichen planus,respectively.No statistical significances of the FI in Rb expression were found between the normal oral mucosa and oral SCC as well as leukoplakia,lichen planus (P> 0.05 ),There was no obvious difference of positive rate of Rb gene expression among the different histological grades of oral SCC(P> 0.05 ).Conclusion ①c-myc oncogene was involved in malignant progression of premalignant lesions.②Negative relationship between the expression of Rb gene and the histological grades of oral SCC.③The alterations of Rb gene and loss of Rb protein may become a potential danger in oral premalignant lesions.

目的 研究C -myc癌基因与Rb抗癌基因在口腔癌的发生及癌前病变恶变过程中的意义。 方法 采用流式免疫荧光技术对 40例口腔鳞状细胞癌 ,10例口腔白斑和 14例口腔扁平苔藓中C -myc和Rb基因蛋白的表达量进行定量研究。结果 C -myc癌基因在口腔癌、白斑、扁平苔藓中表达的阳性率分别为 6 5 %、30 %和2 1%。在C -myc癌基因的平均表达量中 ,口腔癌明显高于白斑和扁平苔藓 ,差异均有显著性 (P <0 .0 5和P <0 .0 0 5 )。Rb抗癌基因在口腔癌、白斑、扁平苔藓中表达的阳性率分别为 30 %、0和 7%。其中口腔癌 ,白斑 ,扁平苔藓中的平均表达量均低于正常口腔粘膜 ,但差异无显著性 (P >0 .0 5 )。口腔癌不同组织学类型之间Rb蛋白的阳性检出率无明显差异 (P >0 .0 5 )。结论 ①C -myc癌基因在口腔癌前病变发展为口腔癌过程中具有重要意义。②Rb基因的表达与口腔癌组织学分级之间无明显关系。③Rb基因表达失活可能成为口腔癌前病变癌变的一种潜在危险
