To investigate the differences of differentiation into dopaminergic (DA)neurons between neural stem cells(NSCs)from fetal rat cerebral cortex and those from midbrain, NSCs isolated from fetal rat cerebral cortex or midbrain were proliferated by the technology of serum-free culturing and single cell cloning, then cells of a single clone were divided into 3 groups and cultured in different media: 10% fetal bovine serum (FBS) in the control group; 10% FBS containing IL-1α in the IL-1α group; 10% FBS containing IL-1α, IL-11, LIF and GDNF in the united factor group, respectively. The DA neurons were indicated by immunocytochemichal staining of tyrosine hydroxylase (TH). The number of TH-positive neurons was counted in 5 randomly selected microscopic visual fields, and image processing was carried out for cellular analysis of cell size and perimeter. At last, a STATA7.0 statistical software was used to analyze the results. In the cortex control group, almost no TH-positive neurons (0.25±0.163) were detected, while some TH-positive neurons (2±0.378) were seen in the midbrain control group. Comparing the number of TH-positive neurons, more were generated in the IL-1α group than in the control group, and as 1.5 times in the midbrain IL-1α group (22.87±1.517) as in the cortex IL-1α group (15.5±0.866). However, there was no significant difference between the number of the TH-positive neurons in the cortex united factor group (25.75±0.940) and that in the midbrain united factor group (28.875±2.125). As far as the cell size and perimeter were concerned, the IL-1α group and the united factor group had an advantage over the control group, and the united factor group exceeded the IL-1α group. These results suggest that neural stem cells derived from midbrain have regional specification to differentiate into DA neurons. However, when induced by IL-1α, IL-11, LIF and GDNF, neural stem cells derived from cerebral cortex can alter its regional specification and differentiate into mature DA neurons.

Objective:To investigate the distribution of astrocytes in the hypothalamus of rats. Methods: Cellular localization of GFAP-labelling cells were mapped in the subdivisions of the hypothalamus of rats with immunohis-tochemistry staining. Results: Dense immunoreactivities of GFAP were found in the periventricular area of the third cerebral ventricle, the supraoptic nucleus, the lateral magnocellular part of the paraventricular hypothalamic nucleus and the median eminence. While only slight GFAP-immunoreactivities was presented in other areas or nuclei of the hypothalamus, such as the anterior area, the lateral area, the lateroanterior nucleus, the suprachi-asmatic nucleus, the arcuate nucleus, the parvocelluar part of the paraventricular nucleus and the ventromedial nuclei i. e. Conclusion: The distribution of astrocytes shows a regional specification characteristic in the hypothalamus of rats, which might be related to some corresponding physiological activities and functions upon these subdivisions.

目的:观察星形胶质细胞在正常大鼠下丘脑的分布。方法:利用免疫组织化学技术,观察胶原纤维酸性蛋白(Glial fibrillary acidic protein,GFAP)在下丘脑的表达。结果:GFAP阳性细胞在第三脑室室周区、视上核、下丘脑室旁核外侧大细胞部、正中隆起大量分布;下丘脑其他区域或核团,如前区、外侧区、前外侧核、视交叉上核、室旁核小细胞部、弓状核、腹内侧核等内分布稀疏。结论:星形胶质细胞在正常大鼠下丘脑的分布有明显区域性,可能与其相应区域的生理活动与调节功能有关。
