By using guinca pig bone marrow as the inductor, changes of induced structures both in quantity and quality were observed when the duration of contact between the inductor and the ectoderm varied. When the time of contact was long enough mainly mesodermal structures of the dorsal side (notochord, muscle) were induced. By using guinca pig bone marrow as the inductor, changes of induced structures both in quantity and quality were observed when the duration of contact between the inductor and the ectoderm varied. When the time of contact was long enough mainly mesodermal structures of the dorsal side (notochord, muscle) were induced. It shifted to lateral and ventral structures (pronephros and blood cells) when the time of contact was shortened. It was assumed that the differentiation of different mesodermal structures might be determined by different quantities of mesodermal inducing-agent entered the ectoderm cells during different durations of contact with the inductor (Tseng, 1963). However, bone marrow may contain more than one inducing-factor, and the results are difficult to be analyzed. It is reasonable to use a more or less pure mesoderm-inducing protein to study the relation between the quantitative differences of the inducing agent and the differentiation of mesodermal structures induced.A mesoderm-inducing protein was extracted from pig liver by phenolpyridine method according to Tiedemann and Tiedemann (1959), and further purified by CM-cellulose column chromatography (Wang, et al., 1963; Bao, et al. 1966). The effective component obtained was homogeneous on free-boundary electrophoresis, but still contaminated with impurities when was analyzed with polyacrylamide gel electrofocusing. A batch of inducingprotein was serially diluted to different concentrations of 600μg/ml, 300μg/ ml, 150μg/ml, 75μg/ml, and 37.5μg/ml. By immersion method a piece of the presumptive ectoderm of early gastrula of the newt, Cynops orientalis, was transfered into 0.08 ml of the inducing-protein solution of different concentrations and treated for 12 hours. The results were summarized in table 1.

Changes in protein synthesis of newt (Cynops orientalis) gastrula ecto-derm after being treated by inducing agents were investigated by O'Farrell's technique of two-dimensional polyacr-ylamide gel elctrophoresis.Proteins we-re seperated according to isoelectric point by isoelectric focucing in the first dimension,and according to mo-lecular size by SDS electrophoresis in the second dimension.The ampho-line compositions used in gel isoelectric focucing were 1.6% pH5-7 and 0.4% pH 3.5-10,gaving a pH range of 4.38-7.15.The discontinuous SDS elec-trophoresis was carried out in 12% gel.More than 300 different compo-nents of proteins in the extract from explants were detected on the slab gel with highly sensitive silver stain. After being induced with the ex-tract from guinea pig bone marrow which had been proved to be a very potent mesodermal inductor,the ex-plants were divided into two groups for continuous cultivation of two daysand five days respectively.The pro-tein patterns in the first group were almost the same as that of normal ectoderm.But the quantity of one protein was increased and of two other proteins were reduced,as shown in Fig 2(a) and Fig 1.The patterns in the second group was quite different.When Fig 2(b) was compared with Fig 2(a),three new proteins appeared (marked by arrows),while the quanti-ties of five other proteins were increased and two reduced. New proteins in the case of neural differentiation two and four days after the treatment of ectoderm explants with Ca~(++)-free medium were not observed on the gel patterns.However,the prominent changes thusfar observed we-re the disappearance of eighteen pro-teins on the gel and the reduction in quantity of other fifteen proteins.The-se were shown in Fig 3(a) and Fig 3(b). These experimental results indica-ted that during the induction of ecto- derm explants,changes of protein pat-tern in the explants have already be-gun before the appearance of distinct morphological characters.These pat-terns were completely different in the case of two different inductive trea-tments which led to either mesodermal or neural differentiations.

用O′Farrell根据蛋白质等电点与分子量的双向电泳法,分析东方蝾螈早期原肠胚外胚层诱导处理后,其蛋白质合成的变化。双向电泳上的蛋白质用银染法显色。结果表明,用豚鼠骨髓抽提液处理后向中胚层分化的外胚层,其蛋白质的图谱在诱导后二天内未见变化,五天出现较明显的变化,有新蛋白的形成。经缺钙处理向神经分化的外胚层,其蛋白质的图谱在处理后的二天和四天都未发现新的蛋白质点,但在4天组中有不少蛋白质浓度下降以及消失。结果说明,诱导分化过程中,在显示明确的形态学特征之前,蛋白质的量与质的变化或多或少就已经开始了,经过不同的诱导处理后,外胚层细胞向中胚层或神经分化过程中,蛋白质的组成是截然不同的。
