Objective:To analyse the correlation of ultrasonographic geometry,oncogenic expression and vasoactive peptide contents in cardiac ventricles and plasma of hypertensive rats.Methods:The ultrasound structural and functional changes,the oncogence c-fos,c-myc expressions,vasoactive peptide(ANP)were detected by echocardiography,immunohistochemistry and radioimmunoassay in cardiac tissue and plasma in 56 renovascular hypertensive rats.Results:The cardiac geometries in hypertensive rats were devided into three patterns such as concentric hypertrophy,eccentric hypertrophy or normal forms,according to the ultrasound parameter changes.The positive expressions of c-myc and c-fos were stronger in concentric or normal patterns than those in eccentric hypertrophy or non-hypertrophic counterparts(P<0.05),following the raise of blood pressure(P<0.05).The ET contents in plasma and cardiac tissue were increased gradually by the order of control group,normal forms,concentric hypertrophy and eccentric hypertrophy,while ANP contents were increased by the order of non-hypertrophy group,eccentric,normal form or concentric groups.Conclusions:There were good correlations among ultrasonic parameters such as diastolic left ventricular posterior wall thichness(LVPWd),diastolic ventricular fiber shortness(FS%),left ventricular mass/body weight(LVM/BW),ejection fraction(EF%)and c-fos or c-myc expression,ET or ANP contents.Both oncogenes and vasoactive peptides may serve as crucial role in developing and progressing cardiac geometric alteration of hypertensive rats.

目的 :探讨高血压发生发展过程中左室构型超声参数、癌基因和血管活性肽表达的特点及其相互关系。方法 :应用高频超声心动图、放免法和 L SAB免疫组化法 ,动态观察肾性高血压大鼠左室重构的超声改变 ,心肌细胞癌基因 c- myc、 c- fos、血浆及心肌组织中血管活性肽内皮素 (ET)、心钠素 (ANP)的含量变化。结果 :高血压大鼠心肌构型变化依据超声参数可分为正常左室构型、向心性肥厚、离心性肥厚等类型。正常构型及向心性肥厚组心肌细胞 c- myc、 c- fos阳性表达率明显高于对照组及离心性肥厚组 (P<0 .0 5 ) ,且 c- myc、 c- fos表达随血压升高逐渐增强 (P<0 .0 5 )。血浆和心肌组织中 ET含量为离心性肥厚组 >向心性肥厚组 >正常构型组 >对照组 ,而 ANP含量则为向心性肥厚组 >正常构型组 >离心性肥厚组 >对照组 (P均 <0 .0 5 )。结论 :L VPWd、IVSd、L VM/ BW、EF%、FS%等超声参数与癌基因、血管活性肽表达有较强相关关系 ,后两者是心脏构型改变的分子生物学基础。

目的 :应用超声心动图评价高血压大鼠心脏结构、功能动态变化 ,同步监测血浆及心肌组织中心钠素(atrial natriuretic peptide,ANP)、内皮素 (endothelin,ET)含量变化 ,分析高血压大鼠心脏结构功能变化与血浆及心肌组织中 ANP、 ET含量变化关系 ,从而进一步了解高血压心脏病左室结构、功能变化的生物化学基础。方法 :SD大鼠左肾动脉狭窄 ,即二肾一夹型 (2 K1C)肾血管性高血压大鼠 (RHR)。术后每周监测尾动脉压及心脏超声学参数。用放免法测定血浆及心肌组织中 ANP、 ET含量。结果 :1、高血压早期 (术后 3周左右 )血浆及心肌组织(左心室 )中 ANP、 ET含量明显升高 ;2、高血压左室向心性肥厚组血浆及心肌组织中 ANP、 ET升高更为显著 ;3、左室离心性肥厚期血浆 ET较向心性肥厚期组更高 ,但心肌组织中 ET含量与其无显著差别 ,而血浆及心肌组织中ANP含量均较向心性肥厚期组低 ;4、相关分析 :(1)血浆及心肌组织中 ANP含量与室间隔厚度 (IVST)、左室后壁厚度 (PWT)、相对室壁厚度 (RWT)呈正相关 ;(2 )血浆及心肌组织中 ET含...
