本实验通过外源性 cAMP 和茶碱对人胃腺癌细胞系 SGC-7901的连续作用,观察到外源性 cAMP 和茶碱对该细胞株的增殖,具有明显的抑制作用。在透射电镜和扫描电镜下,均可见到细胞表面较光滑,微绒毛减少或消失。同时见到细胞膜表面上的 Na~+—K~+—ATP 酶活性受到抑制。本文讨论了外源性 cAMP 对人胃腺癌 SGC-7901细胞系增殖的抑制作用,以及膜表面微绒毛和Na~+—K~+—ATP 酶活性变化的关系。

The wild type cells of human stomach glandular carcinoma, cell line BGC 823, were treated firstly by a chemical carcinogen (MNNG) for the induction of mutagenesis, then the cells were selected in medium with gradually increasing amount of 8-AG (8-azaguanine) from 1-20μg/ml. It was found that the mutant cells could grow vigorously in the medium containing 20μg/ml of 8-AG but not in HAT medium. The HGPRT assay showed an obvious quantitative difference between the wild type and HGPRT mutant cells of BGC 823....

本研究首先用化学突变剂MNNG处理人胃癌细胞系(BGC 823),以提高突变率。然后用递增浓度的鸟便嘌呤类似物8-AG选择次黄嘌呤-鸟便嘌呤磷酸核糖转移酶(HGPRT)缺陷细胞,8-AG递增浓度为1~20μg/ml,历时半年。获得抗8-AG毒性的突变细胞,在HAT培养基中不能生存。经HGPRT酶的同位素法测定,发现野生型与突变型的BGC 823细胞有明显差别。本文还对缺陷型BGC 823的A、B、C、D、E、F株的应用进行了讨论。

The experiment was carried out on cultured human stomach glandular carcinoma (SGC-7901 cell line). We previously reported the cytochemical changes of (Na~+-K~+)-ATPase activity and the changes of microvilli. We here studied the relationship among the cAMP, cAMP-PDEase, Mg~(++)-ATPase activity, the cell growth and differentiation. Cells were exposed to Immol/L cAMP together with 1 mmol/L theophylline. We showed cAMP specific fluorescence by immunocytochemical method, Mg~(++)-ATPase and cAMP-PDEase activity...

The result indicates that exogenous cAMP can inhibit 3', 5'-cAMP-PDEase and Mg~(++)-ATPase activity and increase cAMP level in cell, and cAMP regulated the growth and differentiation of SGC-7901 cell line

本实验通过外源性3′,5′环腺苷酸(cAMP)对人胃腺癌SGC-7901细胞系的连续作用,观察到癌细胞的增殖受到明显抑制的同时,膜表面环腺苷酸-磷酸二酶(cAMP-PDEase)的活性及Mg~(++)-ATPase活性均明显下降,而细胞内cAMP免疫荧光强度则明显增强,进一步证实了细胞内cAMP水平与细胞的生长、分化,及膜表面ATPase、cAMP-PDEase的活性有密切的关系。
