And studies showed that American leafminer began to tunnel leaf from lower leaf segments of plants, there were much more larvea on middle and upper than on lower leaf segments, and most of damaged leaf segments moved up step by step. According to rearing in laboratory and observing in field, we obtained the developmental durations of esch developmental stage of American leafminer on eggplant, pumpkin and kidney bean, respectively. Meanwhile, we observed and described the habits of each developmental stage of it.

The virus is purified by differential and rate-zonal centrifugation,the A260/A280 ratio for purified virus is 1.22. Antiserum which had a homologous titer of 1: 109 determined by ELISA is prepared. The name of zucchini yellow mosaic virus-Xinjiang(ZYMV-XJ)for this virus isolate is proposed.

A new method of microtubular immunofluorescence labeling in plant cells is reported here. After fixation, a stem of pumpkin (Cuturbita moschata) was cut into 100~200 μm sections along its longitudinal axis by hand sectioning. The sections were processed for immunofluorescence labeling and observed under a confocal laser scanning microscope. In this way microtubules were more clearly observed in different cells. 报道一种植物细胞微管骨架免疫荧光定位的新方法。化学固定后,通过徒手切片,将南瓜茎沿纵切面切成100~200μm的薄片,经过免疫荧光定位方法处理,在共焦激光扫描显微镜下,可以对不同类型细胞中微管骨架的排列进行观察。研究发现,南瓜茎细胞内微管骨架排列的主要方式有以下几种:1、网络状微管,多存在于薄壁细胞中,其中微管往往与液泡膜、核膜及质膜相连;2、周质微管,多存在于导管分子或伸长细胞中,其中微管往往彼此平行,并与细胞的纵轴成一定角度;3、纵向微管,存在于筛分子等少数伸长细胞中,其中微管往往接近与细胞纵轴平行。
