Callus induction and plant regeneration were successfully achieved by culturing immature embryos of intergeneric hybrids between Hordeum vulgare (2n=14) and Tri-ticum aestivum (2n=42). Upon repeatedly subculturing the hybrid culture proliferated rapidly, and more than 100 regenerated hybrid plantlets were isolated within six months after inoculation from only a single initial hybrid embryo. The media used for callus initiation, maintenance, shoot differentiatioin and root development were all the same, i.e....

Callus induction and plant regeneration were successfully achieved by culturing immature embryos of intergeneric hybrids between Hordeum vulgare (2n=14) and Tri-ticum aestivum (2n=42). Upon repeatedly subculturing the hybrid culture proliferated rapidly, and more than 100 regenerated hybrid plantlets were isolated within six months after inoculation from only a single initial hybrid embryo. The media used for callus initiation, maintenance, shoot differentiatioin and root development were all the same, i.e. the inorganic ingredients of Murashige and Skoog medium were supplemented with 150 mg L-asparagine and 0.50 mg thiamine-HGl per litre, except that 2,4-D level was reduced step by step from 1.0 to 0.5, 0.1 and 0.0 mg/1. Cytological examinations conformed the hybrid nature of the culture, as well as the culture-derived plants (2m=28). However, mixoploidy was revealed as a distinct feature of them. Apart from the normal dihaploid and aneuploid cells, spontaneous amphiploid cells were also found in the culture and in the root tips of the regenerated plants with a chance of 4 and 0.7 percent respectively. Unfortunately, none of such cells was found in PMCs of the regenerants, suggesting that competition existed between the normal hybrid cells and amphidiploid and aneuploid cells during the course of callus differentiation and ontogenetic development of the regenerated plants. Conspicuous somaclonal variation was observed either among different regenerated plants or among different tillers of the same, plant. Altered characteristics concerned spike morphology, awn length, spike-neck color etc. Variants took up almost one half of the regenerants or about one third of all the culms.

Observations were made on interspecific F1 hybrid plants between two yellow-brown seed lines of B. napus L. and two yellow seed Abissinian mustard varieties ( B. carinata A. Br. )to examine the chromosome number in root tip cells and the meiotic behaviour of pollen mother cells (PMC). For chromosome counting, root meristems were treated with 8-hydroxylquinoline or colchicine for 3-4 hours at room temperature,fixed in the modified Carnoy's solution,colored with acetic carmine,and smeared with. 45% acetic acid. For the observation of meiotic behaviour of PMCs,young floral buds were sampled at 9. 00-11. 00,then treated with Canoy's solution colored with iron haematoxylin and smeared with 45% acetic acid. The results showed that the chromosome number in the root tip cell was 2n = 36 (Fig. la) .which can be a cytological evidence of the hybrid nature of the F1 plants. However,the meiotic behaviour of the PMCs was abnormal. At metaphase I,a number of chromosomes were univalents averaging 16. 06 per PMC. But the average number of bivalents was only 9. 71 per PMC(Fig, 1b-c). The meiotic chromosome configuration was (12-20) Ⅰ +(8-12) Ⅱ +(0-1)Ⅲ (Table 1). There were a few lagged chromosomes formed in anaphase Ⅰ (Fig. ld)and some abnormal behaviour took place at anaphase Ⅱ (Fig. le),which resulted in the degeneration of microspores. As a result,sterile pollen grains were produced (Fig. lf).

对甘蓝型油菜黄褐籽品系881127、Y 173、Y 187与埃芥黄种皮品种:Dodola、216845的种间杂种F_1进行了细胞学研究。结果表明,杂种F_1根尖细胞染色体数2n=36,从细胞学上证实了该种间杂种F_1的杂种性质。杂种F_1花粉母细胞减数分裂行为异常,减数分裂中期Ⅰ形成较多的单价体(平均为16.06条),而配对二价体数目较少,平均只9.71条。染色体基本构型为(12—20)Ⅰ+(8—12)Ⅱ+(0—1)Ⅲ。花粉母细胞减数分裂末期,部分单价体形成落后染色体,导致小孢子退化,产生不育花粉。

花生区组四粒红与根茎区组Arachis glabrata杂交果针经离体培养克服了不亲和性,获得杂种。杂种二代性状出现疯狂分离,但有些性状仍分别倾向双亲。经叶片过氧化物同工酶分析,酶带的条数、宽窄、活性强弱及迁移率,杂种与双亲有明显差异,也有共同之处,初步证明了杂种的真实性。
