Leucoma candida Staudinger and Lymantria dispar(L.)are both major foest pests in Shanxi province. They are distributed widely from northern to southern areas in this province, especially Leueoma candida has caused much damage during recent years. The caterpillars of both species are infected by viruses. The viruses that infected both of the pests usually cause epidemic virosis in natural conditions. It is an important factor in limiting the pest population. It was proved that the viruses which infected both of the pests are nucleic polyhedral viruses(NPV)by negative staining and electron microphotography. The protein coat of(NPV)of Leucoma candida is polyhedral. Their diameters are ranged about 0.94—2.29μ withan average of 1.83μ. The virus particles are rod-shaped. Their diameter and length are about 37×256 mu respectively and there are 2—7 capsomeres in a capsid. The protein coat of(NPV)of Lymantria dispar(L.)is polyhedrae also. Their diameters are about 0.8—3.2μ. The virus partieecs are also rod-shaped. Their diameter and length are about 30×200mμ, and there are 1—12 capsomeres in a capsid.

杨毒蛾和舞毒蛾是我省林业方面主要害虫,从雁北至晋南均广泛分布。特别是杨毒蛾,近年来为害严重,每年成灾面积达百万亩。这两种害虫的幼虫均能受到病毒的感染,它们所感染的病毒在自然情况下,常引起流行病,是仰制种群数量的重要因素。经染色鉴别和电镜观察,这两种害虫的幼虫所感染的均是核多角体病毒。杨毒蛾核多角体病毒包涵体为多角形,大小约为0.94~2.29微米,平均1.83微米,病毒粒子杆状,大小约为37×256毫微米,粒子2~7根成束存在于病毒束内。舞春蛾核对角体病毒包涵体亦为多角形,大小约为0.8~3.2微米,病毒粒子亦呈杆状,大小约为30×290毫微米,粒子1—12根成束存在于病毒束内。

The diameter of virus bundles is 146 ± 37 nm. Single em-bedded viruses (SEV) and multiple-embedded viruses (MEV) coexist in one and the same polyhedron, Individual vitions are rods with an average size of 74 10 × 275 25nm. The size of dissociative virion is 56.3 3.7×465 34.5nm. The size of free virions is considerably smaller than that of the virions embedded in the protein crystal lattice of polyhedrons, the average diameter of the former is only five-seventh as large as the latter. The reduction in size of free virions is eveidently the result of physical and chemical treatments in the progess of specimen preparation of electron microscopy. It seems that the measarements of virus from ultra thin sections would be more reliable than those of free virions. The polyhedral virus has been found by first in China. The polyhedral virus of cyclophragme xichangensis in the rank of classifcation of virus will have waitted to decide.

1981年从罹病的西昌杂毛虫(Cyclophragma xichangensis)幼虫体中,分离到一种包涵体病毒。多角体直径为1.895±0.805μm,病毒束直径为146±37nm。病毒束内有1~10个核衣壳。包埋在多角体内带囊膜的病毒粒子杆状,大小为74±10×275±25nm,经弱减处理而游离的病毒粒子杆状,大小为56.3±3.7×465±34.5nm。游离病毒粒子仅及包埋在多角体蛋白质晶格中病毒粒子的71%,据此认为,观测多角体超薄切片中病毒粒子比观测游离病毒粒子更接近其真实形态大小。西昌杂毛虫多角体病毒,为我国首次发现,国外未见报道。

This paper deals with results of our investigation on the factors affe-cting the propagation of the nuclear polyhedrosis virus (NPV) of the polarlooper,Apocheima cinerarius Erschff. The concentration had a conspicuous effect on the propagation of thenuclear polyhedrosis virus.Among the concentration tested,the 1.1×10~6-1.1×10~7 PIB/ml was found to be the best on early four instars aver-age 2.42-2.64×10~9 PIBs/larvae.When the larvae of poplar looper werefed with the poplar leaves contaminated with polyhedral inclusion bodies(PIB) of the virus,the mortality rate of the infected larvae was depen-ded on concentration of the virus suspension and laral instar. The amount of propagated NPV was found to be proportional to theconcentration of the inoculum suspension.The lag of the death of larvaeinoculated with NPV was influences by the temperature and suspensionconcentration of the polyhedral virus.The propagation of the virus wasinhibited at 35℃ and the optimal temperature was 22—27℃.The NPV would propagaten better in early four-instar larvae than in other instars onan average of 2.42×10~9 PIBs/larvae. The propagation of field indicated that optimum suspension contrationwas 2.5×10~6 PIBs/mu to second-instars and third-instars.Dry proparationof PIB of nuclear polyhedrosis virus of Apocheima cinerarius by precipit-ation with acetone from lactose suspensions indicated that the LC_50 val-ues of 2.84×10~3 PIBs/ml for early third-instar larvae and 95% fiduciallimit was 1.70×10~3-4.96×10~3 PIBs/ml.

为了生产病毒制剂提供大量病原物,本文就影响AciNPV增殖的因素及林间复制技术进行了研究。采用浸叶法接毒感染2—4龄初期幼虫,以便寻找适宜的接毒浓度、虫龄与温度范围。试验证明,接毒的适宜虫龄以4龄初期为最好;接毒浓度以1.1×10~6-1.1×10~7PIB/ml之间为宜,平均幼虫多角体产量为2.42—2.64×10~9PIB/头;增殖的适宜温度在22—27℃之间,温度过高对病毒的增殖不利,为在35℃下增殖则受到抑制。在林间复制时,其喷洒剂量以2.5×10~(10)PIB/亩宜,虫龄以2—3龄占85%左右为好;3—4龄幼虫占85%时,应加大用量1倍。
