The reaction between disulphides and sulphite has been utilized for the determination of disulphide groups in proteins. The sulphydryl groups thus produced were allowed to react with p-chloromercuribenzoate and the increase in light absorption in the ultraviolet region represented the content of disulphide bonds in the proteins examined. Alternatively, the excess of p-chloromercuribenzoate remaining can be extracted into carbon tetrachloride and measured with diphenylthiocarbazone colorimetrically at 625mμ.It...

The reaction between disulphides and sulphite has been utilized for the determination of disulphide groups in proteins. The sulphydryl groups thus produced were allowed to react with p-chloromercuribenzoate and the increase in light absorption in the ultraviolet region represented the content of disulphide bonds in the proteins examined. Alternatively, the excess of p-chloromercuribenzoate remaining can be extracted into carbon tetrachloride and measured with diphenylthiocarbazone colorimetrically at 625mμ.It has also been demonstrated that sulphite combined with p-chloromercuribenzoate to form a complex with a dissociation constant of 6.7mM. Nevertheless, when sulphite was present in excess, the formation of this complex did not affect the accurate determination of disulphide groups in proteins. The interference due to the complex formation between sulphite and p-chloromercuribenzoate can also be eliminated by the addition of a measured amount of hydrogen peroxide to remove the excess sulphite. A series of measurements showed that the experimental error of the direct spectrophotometric determinations in the ultraviolet region was less than 2% whereas that for the diphenyl thiocarbazone method was about 5%.Results of a series of determinations of the content of disulphide bonds of a number of proteins by the above mentioned methods agreed satisfactorily with the values reported in the literature. Moreover, it has also been shown that the reactivity of the disulphide bonds toward sulphite differ markedly for different proteins or even for different disulphide bonds in the same protein. For instance, the disulphide bonds in chymotrypsin and trypsin were readily attacked by sulphite, those in insulin were somewhat more resistant whereas some of the disulphide bonds in ribonuclease or papain required even more drastic conditions before they were quantitatively split by treatment with sulphite.

本文用电子能谱验证了头发角蛋白被H_2O_2等氧化剂氧化时二硫键的主要氧化产物是磺酸基团,从而回答了文献上有关这个问题长期存在的争论。此外文中首次用电子能谱揭示了长发不同部分的表面二硫键被自然氧化的不同情形,如揭示了长发末梢表面竟有50%左右的二硫键被自然氧化,中长发表面也有20~30%的二硫键被自然氧化。本文最后探讨了二硫键氧化降解机理,认为S-S 氧化断裂是主要的,C-S 断裂的可能性也不能完全排除。
