The effects of brain wound tissue extracts on the c-fos proto-oncogene expression and the morphological characters of cultured astrocytes were observed by dot blot hybridization and morphometric method. The results showed that at the first hour after addition of the BWTE to the medium after dissociated primary culture for 7 days, c-fos oncogene expression of the cultured astrocytes was significantly enhanced, compared with the control group. But after two hours, the c-fos gene expression is dramatically decreased....

The results showed that at the first hour after addition of the BWTE to the medium after dissociated primary culture for 7 days, c-fos oncogene expression of the cultured astrocytes was significantly enhanced, compared with the control group. But after two hours, the c-fos gene expression is dramatically decreased. After the addition of BWTE into the culture medium for three days, the morphological changes of newbron rat astrocyte were observed: (1) the cell body was enlarged and became rod-shape from stellate form; (2) the cell processes were increased obviously in diameter, but decreased in the mean number. No obvious multiplication of the astrocyte was observed. These results suggest the injury related factors could invoke c-fos oncogene expression in astrocytes, and reactive astrocytosis in CNS caused by injury factors might be mediated by fos and inn proteins.

本研究应用分子杂交和形态学方法观察脑创伤组织提取液对培养大鼠脑星形胶质细胞c-fos原癌基因表达的影响及形态特征改变。发现在分散培养7d后换置于含有脑创伤组织提取液的培养液30~60min,斑点杂交显示星形胶质细胞c-fos原癌基因表达比加正常脑组织提取液组明显,2h后表达减弱.统计学表明有显著差异.推测损伤因素引起了c-fos原癌基因表达.当将脑创伤组织提取液加入新生鼠星形胶质细胞培养液3d后,体视学观测其细胞数无明显增加.以上结果提示损伤因素诱导星形胶质细胞内表达FOS蛋白可能刺激反应性胶质增生.
