Objective To investigate whether fetal spinal cord (FSC) transplantation effected the expression of the glia fibrillary acidic protein (GFAP) mRNA and immunoreaction of GFAP as well as functional recovery after spinal cord injury in rats. Methods Experimental rats were divided into 2 groups, spinal cord hemisection injury combination with transplantation of FSC to injured site group (Group A), spinal cord injury and recept of gelfoam pledgets group (Group B). Rats were killed at 1, 3, 7, 14, 28 days. Then, open field and inclined plane tests, somatosensory evoked potentials(SEP) and motor evoked potentials(MEP) were examined. An in situ hybridization and immunocytochemistry was used to examine the spinal cord lesion and neighbor areas. The positive cells were quantitatively analysed by a computer image analysis system. Results In Group A, the number of cell expression protein GFAP mRNA and GFAP immunoreaction was more than that in Group B. By 28 days following injury, positive astrocytes cell of Group A returned to normal level but, the positive cells increased by 14 days postinjury in Group B. These increases in GFAP positive cells were paralleled by a significant improvement in neurological function recovery. Conclusions FSC transplantation is sufficient to reduce multiple aspects of gliosis and can promote neurological function recovery of adult rats after spinal cord injury.

目的 研究大鼠脊髓损伤后胚胎脊髓 (FSC)移植是否能够影响胶质纤维酸性蛋白(GFAP)的表达和伤后大鼠后肢功能的恢复情况。 方法 将 6 0只大鼠分为脊髓半切洞损伤 +胚胎脊髓移植组 (A组 )和单纯脊髓半切洞损伤组 (B组 )。伤后 1,3 ,7,14,2 8d ,应用行为学和电生理检查观察大鼠功能恢复情况 ,应用原位杂交和免疫细胞化学方法观察GFAP的表达 ,并采用计算机图像分析技术 ,进行定量分析。 结果 大鼠脊髓损伤后GFAP的表达A组明显高于B组 ,A组损伤脊髓高表达GFAP持续至伤后 2 8d ,B组仅持续至伤后 14d。增加的GFAP阳性细胞数目与神经功能的改善相一致。 结论 胚胎脊髓移植能诱导大鼠损伤脊髓高表达GFAP免疫反应和GFAPmRNA蛋白 ,并促进大鼠脊髓功能恢复

Objective To investigate whether fetal spinal cord (FSC) transplantation affects the expression of the neuronal growth-associated protein (GAP-43) as well as functional recovery after hemisection spinal cord injury in the rats. Methods Sixty-six adult Wistar rats were randomly divided into three groups: spinal cord hemisection injury combined with transplants of FSC to injured site (transplantation group A, n=30), spinal cord injury received gelfoam pledgets group(hemisection group B, n=30) and control group (n=6). After operation open field and inclined plane tests were performed to test the function of the rear limbs. The animals were sacrificed 1, 3, 7, 14 and 28 day after surgery. The spinal cord lesion and the neighborhood areas were examined by in situ hybridization. The GAP-43 mRNA positive cells were quantitatively analyzed by a computer image analysis system. Results The GAP-43 mRNA positive expression cells were detected in the gray matter one day after spinal cord injury(SCI). The expression of GAP-43 mRNA and protein cells increased rapidly after operation and reached their peak at day 7 after SCI in both group A and B; the number of neurons expressing GAP-43 mRNA approximately doubled in the rats received fetal spinal cord graft in transplantation group(39.24± 6.83) compared with that in hemisection group (21.48± 7.83, P< 0.05). The positive cells in transplantation group maintained at high level to day 28; however, in the hemisection group the positive cells decreased significantly at 14 days. The transplantation group had a significant higher figure than that in the hemisection group. The increase in GAP-43 mRNA positive cells was paralleled by a significant improvement in neurological function evaluated by open-field and inclined plane tests. Conclusion Fetal spinal cord graft can induce a high level in GAP-43 mRNA expression which can improve recovery of function after SCI of rats.

Objective To study the statement of immunoreaction of c Jun and functional recovery of spinal cord injury in rats after fetal spinal cord(FSC) transplantation.Methods Experimental rats were divided into two groups:spinal cord hemisection injury combination with transplants of FSC to injured site group(group A),and spinal cord injury that received gelfoam pledgets group(group B).On the lst,3th,7th,14th and 28th day after operation,6 animals in each group at each time phase,were observed for their function recovery with the methods of praxiology and electro physiological examination,the animals were then sacrificed for the observation of the statement of c Jun reaction by immnochemistry method and computer image analysis system was used for quantitative analysis.Results The c Jun immunoreaction of group A were significantly higher than that of group B.On the 28th day after injury,positive cells of group B could still be recognized and that of group A returned to normal level at 7 days after operation.These increases of GFAP positive cells were paralleled with a significant improvement in neurological function recovery.Conclusion FSC transplantation is sufficient to increase the statement of immunoreaction of c Jun and promote the recovery of neurological function of rats after spinal cord injury.

目的 研究大鼠胚胎脊髓移植后能否影响c Jun免疫反应的表达和脊髓损伤后大鼠功能的恢复。方法 将动物分为脊髓半切洞损伤加胚胎脊髓移植组 (A组 )和单纯脊髓板切洞损伤加明胶海绵填塞组 (B组 )。每个时相点每组 6只动物 ,术后 1,3,7,14和 2 8天 ,应用行为学和电生理检查观察大鼠功能恢复情况 ,应用免疫细胞化学方法观察c Jun免疫反应的表达 ,采用计算机图像分析技术 ,进行定量分析。结果 大鼠脊髓损伤后c Jun免疫反应的表达A组明显高于B组 ,胚胎脊髓抑制后可使损伤脊髓高表达c Jun免疫反应持续到术后 7天。增加的c Jun免疫反应阳性细胞数目与神经功能的改善相平行。结论 胚胎脊髓移植后可使损伤脊髓高表达c Jun ,并促进大鼠功能恢复。
