目的:研究瑞香素的抗溶血与抗红细胞膜脂质过氧化作用。方法:用常规的溶血毒性体外检测法及高效液相层析定量测定脂质过氧化产物法,检测不同浓度瑞香素对伯氨喹(PQ)溶血毒代谢产物5,6-二羟基8-氨基喹啉(AQD)诱发溶血与红细胞膜脂质过氧化的抑制率。结果:10至80μmol/L瑞香素对AQD溶血毒性与红细胞膜脂质过氧化的抑制率分别为33.0%-69.2%和11.9%-58.2%,且呈一定的剂量-反应关系。结论:瑞香素具有抗溶血与抗红细胞膜脂质过氧化作用。

Purpose To estabish a conventional method for generating complement-depleted sera and to test the inhibitory effects of Chinese traditional herb,Eucommia ulmoides Oliver,toward complement system and relevant mechanisms. Methods ① Different complement component depleted sera were prepared by co-incubating the designated complement antibodies with sera. ② Anti-hemolytic effects of the herb extracts were determined by using classical pathway assay.③ The anti-hemolytic mechanism from one of the herb extractions was systematically evaluated by using the different complement depleted-sera. Results ① The sera depleted of C3,C4 and C5 were successfully prepared in our laboratory,and their effects were a bit better than the commercial available products.② The extraction from Chinese traditional herb,Eucommia ulmoides Oliver,showed moderate inhibition toward the complement system. Conclusions The Chinese traditional medicine Eucommia ulmoides Oliver has an inhibitory effect on the complement.Maybe C3,C4 are the targets for Eucommia ulmoides Oliver anti-complement action,however C5 maybe not.

目的建立方便快速的补体缺失血清制备方法,利用补体缺失血清,研究杜仲抑制补体系统的作用及其机制。方法①将指定补体抗体与相应补体结合,制备不同补体成分缺失血清;②通过补体经典激活途径,测定中药及其提取物的抗溶血作用;③以不同成分缺失的补体缺失血清,评估中药及其提取物对不同补体成分的影响。结果①成功制备补体C3、C4、C5缺失血清,略优于市售成品;②杜仲树皮部位有抑制补体溶血作用。结论杜仲树皮部位有一定抑制补体溶血作用,其抑制补体溶血的位点可能位于补体C3、C4段上,而不在C5段上。

Objective: To study the hemolytic and anti-hemolytic activities of individual ginsenosides through the experiments of hemolytic activity or anti-hemolytic activity measurements. Methods: Measure the absorbance of several samples which consist of different concentration of individual ginsenosides and 1% erythrocyte solution, some also contain Saponin(SIGMA), set up the simulation-relationship-curve between the concentration of individual ginsenosides and the hemolytic activity or the anti-hemolytic activity by statistical software, give 50% hemolysis (HD50) and 50% anti-hemolysis (P50) through data analysis.Results: The individual ginsenosides measured in the experiment have the properties of hemolytic activity and anti-hemolytic activity respectively or both. Their abilities of their hemolysis or anti-hemolysis are different from each other. Conclusion: Protopanaxtriol-type ginsenoside-Rg1,-Re showed anti-hemolytic activity, whereas-20(S)Rg2,-20(R)-Rg2 and Rh1 exhibited both activities in different concentration; Protopanaxodiol-type ginsenoside-Rb1,-Rb2,-Rc and Rd showed anti-hemolytic activity, however Rd also showed hemolytic activity in a higher concentration solution; Oleamolic acid type ginsenoside-Ro exhibited anti-hemolytic activity only.

目的通过对几种单体人参皂苷溶血、抗溶血作用进行研究,得到人参皂苷浓度与溶血度间的关系曲线,探讨其作用规律。方法不同浓度的单体人参皂苷溶液和相同浓度的兔血红细胞悬浊液组成的混合溶液充分作用后,用比色法测定溶血度;不同浓度的单体人参皂苷溶液与相同浓度的对照皂苷溶液混合后加入相同浓度的兔血红细胞悬浊液,三者组成的混合溶液充分作用后,用比色法测定溶血度并计算保护溶血百分数。通过数理统计软件对数据进行分析并拟合人参皂苷溶血或保护溶血作用的曲线。结果在测定的人参皂苷各单体中分别或兼具有溶血和抗溶血的特性,且溶血和抗溶血能力各不相同。结论原人参皂苷三醇型人参皂苷Re、Rg1、20(R)-Rg2、20(S)-Rg2和Rh1都具有抗溶血作用,其中20(R)-Rg2、20(S)-Rg2和Rh1在较高浓度时还表现出溶血作用;原人参皂苷二醇型人参皂苷Rb1、Rb2、Rc和Rd均表现出抗溶血作用,其中Rd还在浓度较高时表现出了溶血作用。齐墩果酸型人参皂苷Ro在较低浓度便表现出抗溶血作用,未见溶血作用。
