In the 3'-MeDAB induced liver tumour, it was found that the activities of glucoses-6-phosphate dehydrogenase, 6-phosphogluconic dehydrogenase and dipeptidases were higher than those in the normal liver, while the activities of glutamic dehydrogenase, glutaminase(phosphate-activated), ornithine carbamyl transferase, tryptophan pyrrolase, threonine dehydrase and tyrosine transaminase were lower or even absent.Changes in most enzyme activities were observed in the precancerous stage and the pattern of these changes...

In the 3'-MeDAB induced liver tumour, it was found that the activities of glucoses-6-phosphate dehydrogenase, 6-phosphogluconic dehydrogenase and dipeptidases were higher than those in the normal liver, while the activities of glutamic dehydrogenase, glutaminase(phosphate-activated), ornithine carbamyl transferase, tryptophan pyrrolase, threonine dehydrase and tyrosine transaminase were lower or even absent.Changes in most enzyme activities were observed in the precancerous stage and the pattern of these changes followed that in the liver tumour. Thus in the course of carcinogenesis, the activities of glucose-6-phosphate dehydrogenase, 6-phosphogluconic dehydrogenase and dipeptidase(glycylglycine as substrate)increased while those of glutamic dehydrogenase, glutaminase and ornithine carbamyl transferase decreased. However, there was no significant change in the activity of dipeptidase when DL-alanylglycine was used as substrate. Changes in activity of tryptophan pyrrolase and tyrosine transaminase were not pronounced as compared with those of the control. The activity of glutathione reductase in the liver tumour was similar to that of normal liver, but it increased from the 4th to 13th week of feeding of the carcinogen. Threonine dehydrase was remarkably influenced by the nutritive factor of the basal diet so that the effect of 3'-MeDAB on threonine dehydrase could not be observed during carcinogenesis.A comparative study has been made with 2-MeDAB, a non-carcinogenic substance. It had no apparent effect on the above enzymes except that it caused the activity of glutamic dehydrogenase to be higher than that of the control.Further experiments have shown that the specific activities of glutamic dehydrogenase and glutaminase in the mitochondria of liver tumour were lower than those of the control. In the precancerous liver the specific activity of glutaminase in the mitochondria was lower than that of the control, while there was no significant change in the case of glutamic dehydrogenase.From these results together with those from other laboratories a possible biochemical mechanism of the carcinogenesis induced by 3'-MeDAB was proposed(Fig. 12). It was suggested that the change in enzyme activity during carcinogenesis may possibly have resulted from the carcinogen being first metabolized in the liver, leading to a higher activity of the oxidative metabolism of glucose-6-phosphate and exhibiting influences on liver enzymes by the metabolites of 3'-MeDAB through different mechanisms. Consequently there produced an abnormal growth and differentiation of liver cell which became neoplastic with the formation of liver cancer, 2-MeDAB may be metabolized in a different way from that of 3'-MeDAB thus producing different effects on most enzyme activities. It is therefore evident that the specific effect of 3'-MeDAB on the liver enzymesmay be closely related to its property of carcinogenisity.

Plant indican (indoxyl-β-glucoside) is identified as the principal constituent in the extract of Ta-Tsing-Yeh (大青叶), leaves of Polygonum tinctoria Lour., a kind of medicinal herb used for centuries. A preliminary test recently made in this laboratory has disclosed that indican possesses some anti-viral activity. Oral doses of 150-200 mg of plant indican were given to human subjects, including normal man and hepatitis patients, as well as dogs and rabbits. The 24-hour urine was collected and treated with Dalgiesh's deactive carbon method, then paper chromatographed and the metabolites identified. Beside unchanged plant indican, urinary indican, and indoxyl-d-glucuronic acid which were supposed to be the main metabolites, were found in all samples of urine. Furthermore, some strongly polar unknown compounds were also found which we presumed to be x-hydroxy indoxyl conjugates. Anthranilic acid and O-aminobenzoylglycine were detected as transformation products in the urine of the rabbit only. The metabolic products found in the urine samples of both normal persons and patients of hepatitis revealed no qualitative difference. The results indicates that the bio transformation of plant indican is in accord with the normal route of physiological detoxification. No attempt on quatitative determinations was made except some simple comparisons. One more point worthy of mentioning is that, according to Tollens and Kauffmann simple indole (200 mg) taken by human beings is verted to urinary indican only and no indixyl-d-glucuronic acid formed.

In a previous communication, we reported that after intraperitoneal injection of N-C~(14)-formylsarcolysine to normal rats, about 54.3% of the radioactivity was excreted in the urine in five hours. In the present paper, the isolation and identification of three metabolites of N-C~(14)-formylsarcolysine in rat and mouse urine are described. In a previous communication, we reported that after intraperitoneal injection of N-C~(14)-formylsarcolysine to normal rats, about 54.3% of the radioactivity was excreted in the urine in five hours. In the present paper, the isolation and identification of three metabolites of N-C~(14)-formylsarcolysine in rat and mouse urine are described. Paper chromatograms (using n-butanol: acetic acid: water, 5:1:4, as the developing solvent) of urine from normal rats given N-C~(14)-formylsarcolysine usually showed three radioactive spots with Rf values of 0.86 (metabolite 1), 0.45 (metabolite 2) and 0.13 (metabolite 3). Both metabolite 1 and metabolite 2 were found to absorb ultraviolet light at 260mμ. Metabolite 1 accounted for about 52.7% of the total radioactivity in the urine and was believed to be unchanged N-C~(14)-formylsarcolysine, since they behaved similarly on paper chromatograms. Metabolite 2 accounted for about 20.2% of the urinary radioactivity. The Rf value and the ultraviolet absorption spectrum of an alcohol eluate of the spot were similar to or identical with that of the hydroxyl form of N-formylsarcolysine. This may imply that metabolite 2 is p-di (β-hydroxyethyl) amino-N-formylphenylalanine. Since metabolite 3 was shown to be unable to absorb ultraviolet light and was found to have an Rf similar to formic acid, it is likely that this metabolite is formic acid. These three metabolites were also found in the urine of Yoshida solid carcinoma rats, Krebs-2 ascites carcinoma mice and spindle cell sarcoma B_(22) mice. It is unlikely that sarcolysine is an important metabolite of N-formylsarcolysine since no ultraviolet absorbing spot was detected which behaved as authentic sarcolysine. However, the possibility that N-formylsarcolysine was converted to p—di (β-hydroxyethyl) aminophenyl alanine through sarcolysine should not be excluded.

正常大鼠或吉田肉瘤大鼠腹腔注射N[C~(14)]-甲酰溶肉瘤素后第一次所排尿中,最少含有三个代謝产物。代謝物1为未变的N[C~(14)]-甲酰溶肉瘤素,其含量为尿中放射性的51.5—52.7%。代謝物2为未脫掉甲酰基的含羥基化合物排出量約为尿中放射性的20.2—23.1%。代謝物3可能是C~(14)-甲酸,排出量約为尿中放射性的9.5—12.6%。自梭形細胞肉瘤B_(22)及Krebs-2腹水癌小鼠尿中亦得到相似的結果。溶肉瘤素并非N[C~(14)]-甲酰溶肉瘤素的主要代謝产物,但并不能排除N[C~(14)]-甲酰溶肉瘤素通过溶肉瘤素轉变为溶肉瘤素羟基水解物的可能性。 N[C~(14)]-甲酰溶肉瘤素在正常动物体內的代謝在质与量上与肿瘤动物者并无不同;在对N-甲敏感瘤动物(吉田肉瘤大员、Krebs-2腹水癌小员)与不敏感瘤动物(梭形細胞肉瘤B_(22)小鼠)之間亦无明显差异。
