The viaointy of cryopreserved bone marrow cells used for autologous bone marrow transplantation were studied.1. The CFU_(GM) colony forming ability of normal bone marrow cells was estimated by three different culture methods. 2. Normal bone marrow samples from 11 individuals were cryopreserved in liquid nitrogen. The nucleated cell recovery rate, trypan blue exclusion rate and CFU_(GM) recovery rate were studied after different intervals of storage. It appears that the viability of CFU_(GM) did not change much even after 50 weeks of storage. 3. After a two-step centrifugation, approximately 80±2.6% of nucleated cells could be seperated. 4. A first case of clinical trail was also reported. After one month of cryopreservation, the nucleated marrow cells from a patient with cervical carcinoma were infused back. The total infused nucleated cell count was 0.53×10~(10) and the total infused CFU_(GM) was 0.27×10~6. No untoward reactions are found after autotrasfusion.

本文旨在研究自体骨髓移植中多量骨髓的冷冻保存问题。(1)比较了正常骨髓粒系定向造血干细胞(CFU_(GM))活力的三种测试方法。在体内扩散盒培养方法中,用环磷酰胺0.3mg/g代替照射处理小鼠。(2)11例正常骨髓标本冻存于液氮并定期复温观察有核细胞回收率、锥蓝拒染率及CFU_(GM)回收率。当骨髓标本长期冻存达50周时,上述指标未见明显下降。(3)用二步离心法浓集多量骨髓的有核细胞。有核细胞可回收82.5±2.6%,所得有核细胞体积仅为原骨髓体积的三分之一左右。(4)在上述实验研究基础上,本文冻存了一例宫颈癌患者的自体骨髓。一个月后回输给患者的有核细胞总数为0.53×10~(10),CFU_(GM)总数为0.27×10~6。输注后患者的情况良好。

Bone marrow is drawn out before chemotherapy, preserved in vitro for some time, and after the effects of the medicine disappear, retransfused into the body. This is what autologous bone marrow transplantation ( ABMT ) means. Thus the above-mentioned harmful effects can be avoided. Therefore, whether or not we can effectively preserve cells and sustain its viability and function is indeed -the key to the successful use of ABMT. The article introduces the cryopreservation of bone marrow. The result attained is very satisfactory. The bone marrow of human beings is effectively preserved for three davs at 4℃. Recoverable rate of the nuclear, exelusion of trypon blue and colony-forming-unit culture ( CFU-C ) are 84%, 91% ar.d 97% respectively. When the bone marrow is preserved in liquid nitrogen, the viability and function of the cells can be sustained much longer. The recoverable rate of the nuclear cells, exclusion of trypon blue, DNA and CFU-C arc 76%, 89%, 77% and 79% respectively. Successful preservation of hematoiogic stem-cell in vitro provides predisposing factor fcr applying ABMT clinically.

在恶性肿瘤的治疗中,国外目前应用强烈化疗或放疗,以期达到根治之目的.但其严重的骨髓抑制大大地限制了它的应用.自身骨髓移植就是在化疗前从人体内抽出,并在体外保存一段时间,然后待药物的毒性作用消失再将其回输入体内.这就避免了上述的副作用.因此能否有效地保存细胞,维持其活力和功能是自身骨髓移植的关键.本文介绍了骨髓的保存方法,尤其是超低温保存.取得的结果是令人满意的.人骨髓于4℃可有效地保存3天,有核细胞、锥蓝拒染率和粒系定向干细胞(CFU—C) 的回收率分别为84%、91%和97%.骨髓贮存于液氮中,其细胞的活力和功能得以较长保存,有核细胞、锥蓝拒染率、脱氧核糖核酸(DNA)和CFU—C的回收率分别为76%、89%、77%和79%.骨髓造血干细胞在体外成功的保存,为临床开展自身骨髓移植提供了先决条件.

In an effort to evaluate the possible effect of cryopreserved autologous bone marrow infusions in man,11 patients with cancer were treated with high-dose chemotherapy or large-field radiotherapy followed by an infusion of cryopreserved autologous bone marrow harvested before the treatment. In an effort to evaluate the possible effect of cryopreserved autologous bone marrow infusions in man,11 patients with cancer were treated with high-dose chemotherapy or large-field radiotherapy followed by an infusion of cryopreserved autologous bone marrow harvested before the treatment. The result indicated that cryopreserved autologous bone marrow infusions hastened hematopoietic recovery in the patients who had received the above mentioned treatment, and the effect could be beneficial to the patients. The methods of harvesting autologous bone marrow cryopreservation and determination of cell vitality were described and discussed. It may be concluded that the procedure is safe for clinical trial. It seems to be a promising approach that may offer hope to cancer patients who have failed to respond to conventional therapeutic modalities.

本文报道了11例恶性肿瘤患者,大剂量化疗或大面积(全身)放射治疗后,使用冷冻保存的自身骨髓对加速造血功能恢复的初步效果。回输冷冻保存的自身骨髓,对受药物或射线抑制的造血系统有促进恢复和重建造血功能之效应。讨论了14例患者自身骨髓采取、冷冻保存和活性测定的方法,证明临床使用是安全可靠的。自身骨髓输注,对常规治疗无效的患者提供了新的治疗途径,但尚处于试验阶段,需累积更多的病例资料才能加以肯定。
