本文探讨了应用DAPI(4’6’-二脒基-2-苯基吲哚)荧光技术对冷冻胚胎移植前进行活力鉴定的实用性。将406个晚期桑椹-早期囊胚阶段的小鼠冷冻胚胎分8批解冻后按形态鉴定标准分为A、B、C、D级。各级胚胎随机分为常规形态鉴定组(平行对照组)和形态-荧光鉴定组(自身对照组)。试验组各级胚胎经DAPI染色后按荧光鉴定标准分为Ⅰ、Ⅱ、Ⅲ、Ⅳ级。通过体外培养对鉴定结果进行验证。试验组224个胚胎的培养结果表明:在胚胎存活率、膨胀率和孵化率方面,荧光法的Ⅰ和Ⅱ级均优于形态学方法的A和B级,两种方法中存活率相同的胚胎级别(荧光法Ⅰ和Ⅱ级;形态学A级)中的活胚检出率,荧光法为94.7%,形态学方法为68.7%,差异极为显著。试验组各批胚胎之间质量上存在显著差异。这种差异对荧光鉴定结果影响较小,而对形态鉴定结果影响显著。通过体外培养和部分绵羊及小鼠冷冻胚胎的移植试验,证明荧光鉴定不影响胚胎在体外和体内的正常发育。试验结果表明:改进的DA-PI荧光鉴定法具有准确客观、快速简便、安全可靠的优点,进一步提高了冷冻胚胎活力鉴定的技术水平和活胚利用率。

In this paper, the binding mechanism of 4'6-diamidino- 2 -phenylindole (DAPI)with DNA is studied by the means of Raman spectroscopy, It is believed that DAPI binds with DNA in a non-inlaid way to form hydrogen bomds with AT base pairs in the minor grooves of DNA double helix, and the binding is inhibi-ted at low pH value and high salt concentration also makes it difficult.

本文以小牛胸腺DNA和λDNA为材料,首次用激光拉曼光谱研究了4’6二脒基—2—苯基吲哚(DAPI)与DNA的结合机制.认为DAPI是以非镶嵌的方式在DNA小沟中同DNA的AT碱基对此氢键结合.在低PH下,这种结合受到抑制,盐浓度的高低与DAPI-DNA复合体的荧光强度成负相关.
