Genes of a novel adult diarrhea rotavirus(NADRV) strain J19 were amplified by an improved single primer sequence-independent amplification method,cloned into pMD18-T vector,sequenced and analyzed.Genomic segments 2,4 encoding VP2,VP3 of NADRV-J19 were 2969bp,2204bp in length,and the deduced proteins were 973aa and 719aa,respectively.Identity of VP2 of J19 to human group B rotavirus strain IDIR was 47.2 % and that of VP3 of J19 to human group C rotavirus strain Cowden was 25.1%.Phylogenetic analysis of VP2 showed that NADRV-J19 positioned closely to outgroup and the ancestral nodes of group A,B and C lineages, although with a preferred association with group B lineage,which corresponded to the phylogenetic analysis of VP6.The above results allowed us to deduce a conclusion that J19 would be a strain closely related to the evolution and origin of group B rotaviruses and they also indicated that VP2 would be valuable for studying the evolution and origin of rotaviruses.This study is the first report on sequence analysis of genes of NADRV-J19 encoding VP2 and VP3.

Objective For better understanding of the molecular characteristics of the gene of a novel adult diarrhea rotavirus strain J19 encoding a UNA-dependent RNA polymerase. Methods The VP1 gene of NADRV-J19 was amplified by an improved single primer sequence-independent amplification method, cloned into pMD18-T vector, and then sequenced. This gene was analysed by the software DNAStar and PHYLIP and the phy-logenetic tree was drawn. Results The gene 1 of NADRV-J19 encoding VP1 contained 3538 bp, which encoded a predicted protein of 1167 amino acids. The identity of VP1 of NADRV-J19 to group B rotavirus strain IDIR was 55.7%; identities of VP1 of NADRV-J19 to group A rotavirus strain SA11 and C rotavirus strain Bristol were 20.4% and 19.2% respectively. Phylogenetic analysis of VP1 of NADRV-J19 showed that NADRV-J19 clustered close to out group and the ancestral nodes of group A, B, and C lineages, although with a preferred association with group B lineages. Conclusion VP1 of NDRV-J19 was different from those of other known rotaviruses. A novel adult diarrhea rotavirus strain J19 would be a strain related tightly to the evolution and origin of group B rotaviruses.

目的进一步了解新的成人腹泻轮状病毒J19株依赖RNA的RNA聚合酶基因特征。方法利用一种改良的非依赖核酸序列的单引物扩增方法扩增新的成人腹泻轮状病毒J19株的VP1基因,克隆至pMD18-T中并进行测序。利用DNAStar和PHYLIP软件包进行序列之间的比较分析并绘制系统发生树。结果J19株的VP1基因为基因1,全长3538个核苷酸,编码1167个氨基酸。氨基酸序列同源性分析表明J19株的VP1蛋白序列对B组轮状病毒IDIR株的一致性为55.7%,对A组轮状病毒SA11株和C组轮状病毒Bristol株的一致性分别为20.4%、19.2%。对J19株的VP1蛋白序列的遗传进化分析表明,J19株在系统发生树上的位置是靠近外群蛋白并靠近A、B和C组轮状病毒分支的根部,而且它比较偏向于B组轮状病毒的分支。结论J19株的VP1蛋白序列与其他轮状病毒的VP1蛋白序列存在显著差异。J19株可能是一个与B组轮状病毒的起源和进化密切相关的毒株之一。

Objective To amplify large genome segments of a novel adult diarrhea rotavirus strain J19. Methods Using a novel adult diarrhea rotavirus strain J19 (NADRV-J19) as an example, 7 small genes of NADRV-J19 were cloned and sequenced by using the single-primer sequence-independent strategy; seven pairs of inhibitory primers from gene 5 to gene 11 were used to prevent amplification of their corresponding genes and improve amplification of large genome segments in one-tube polymerase chain reaction(PCR). Results When two pairs of inhibitory primers were used to inhibit amplification of two corresponding small genes, yields of PCR products for large genome segments more than 2 kb were significantly improved in one-tube PCR. PCR products of genes were cloned into pMD18-T vector, and sequenced. Finally, 3 full-length cDNA clones for gene 2, 3, and 4 of J19 were obtained. Conclusion The improved single primer sequence-independent method could be used to clone large genome segments for rotaviruses.

目的扩增、克隆新的成人腹泻轮状病毒J19株的大片段基因。方法以新成人腹泻轮状病毒J19株的病毒核酸为材料,利用非依赖核酸序列的单引物扩增方法,扩增新的成人腹泻轮状病毒J19株的第5-11基因;根据第5-11基因序列设计7对抑制性引物,通过在PCR扩增体系中添加小片段基因的抑制性引物来提高大片段基因的PCR产量。结果当在PCR反应体系中添加2对抑制性引物时,大于2kd的大片段基因的PCR产量得到显著提高。将扩增的基因片段克隆至pMD18- T后进行测序,最后得到J19株第2、3、4基因的全长eDNA克隆。结论这种改良的非依赖核酸序列的单引物扩增方法可以用于对轮状病毒大片段基因的扩增和克隆。
