从我国新发生的一种家兔急性败血性传染病死兔内脏抽提物中,观察到典型的病毒粒子,回归兔可引起典型发病,再从病死兔内脏回收到同样病毒,证明该病系病毒性传染病,暂定名为“兔病毒性出血症”,病原暂定为“兔出血症病毒”。经初步鉴定,认为本病毒可能是一种首次发现的新病毒,属双股RNA病毒。但从病毒大小和核酸节段看,又不同于呼肠病毒科。最终归属正在进一步研究。

After the contaminating nonviral nucleic acid was degraded by nuclease, the virus nucleic acid was extracted and further purified by agarose gel electrophoresis-electroelutiou. Rabbit hemorrhagic disease virus (RHDY) was purified by PEG sedimentations, organic solvent extraction and differential centrifugation. After the contaminating nonviral nucleic acid was degraded by nuclease, the virus nucleic acid was extracted and further purified by agarose gel electrophoresis-electroelutiou. The purified nucleic acid was hydrolysed by formic acid to its component bases, which were determined by highperformance liquid chromatography (HPLC). The base composition was as follows: C 23.7%; G 20.5%; T 31.9%; A 24%. The G+C mol% was 44.2.

本试验应用聚乙二醇(PEG)沉淀、有机溶剂抽提和差速离心等方法,提取和纯化兔出血症病毒。用核酸酶降解病毒子表面污染的核酸后,再抽提,得到病毒核酸,并经琼脂搪凝胶电泳-电洗纯化,得到高纯度的病毒核酸。应用高效液相色谱对提纯的病毒核酸分析结果表明,兔出血症病毒的核酸为ssDNA.它的碱基组成为:C23.7,G20.5,T31.9,A 24.0。它的G+Cmol%为44.2。
