All trans retinoic acid(ATRA), 13-cis retinoic acid (CRA) and vitamin A (Vit A)were investigated in the plasma, leukocytes and erythrocytes from leukemia patients and healthy individuals by reverse phase high pressure liquid chromatography(HPLC). The following results were obtained. These data suggested that there is probably abnormal retinoic acid metabolism in leukocytes from leukemic patients, and this abnormality may be responsible for the leukemogenesis.

采用高效液相色谱法研究 5 6例孔源性视网膜脱离患者的视网膜下液 (SRF)及血清维A酸的含量 ,并观察部分患者服用维生素A对其的影响。结果示 ,患者SRF中维A酸的含量与增殖性玻璃体视网膜病变 (PVR)程度、视网膜脱离范围和术前服用维生素A的天数呈正相关 (r1=0 .85 1,P <0 .0 0 ;r2 =0 .4 5 2 ,P <0 .0 1;r3=0 .4 3 1,P <0 .0 5 ) ;血清维A酸的含量与眼部病变无关 ;SRF中维A酸在口服维生素服药第 4d时显著增高。结果表明视网膜脱离后 ,色素上皮和神经上皮之间代谢关系发生紊乱 ,造成维A酸循环利用障碍。服用维生素A有利于阻止PVR的发生和发展。

目的探讨细胞内细胞色素氧化酶 p45 0 (CYP)表达、视黄酸 (RA)代谢与细胞凋亡的相互关系。方法采用脂质体介导的质粒转染和逆转录 - PCR等实验技术方法做有关分析检测。结果野生型 HL- 6 0细胞 hp45 0 RAI表达微弱 ,脂质体介导的 hp45 0 RAI质粒转染可使 HL- 6 0细胞稳定表达 hp45 0 RAI;hp45 0 RAI转染对 ATRA诱导的 HL- 6 0细胞增殖能力无明显影响 ;野生型 HL- 6 0细胞有较强 Bcl- 2表达 ,ATRA处理抑制未转染细胞 Bcl- 2表达 ,但单纯 hp45 0 RAI转染、hp45 0 RAI转染 +CYP抑制剂或加用 IFN- α均能增强 Bcl- 2表达 ,其中以加用 ketoconazole作用更为显著。结论 RA敏感 HL- 6 0细胞中影响 RA代谢的主要 CYP并非 hp45 0 RAI,hp45 0 RAI转染能增强 Bcl- 2表达 ,但对 HL - 6 0细胞增殖能力无明显影响 ,CYP抑制剂、IFN-α可能通过抑制细胞内 RA代谢 ,而增加 HL - 6 0细胞Βcl- 2表达。
