Nimodipine as a calcium channel antigonist, bas been used in the treatment of acute brain iechemia. The article was oberved the contents of Nitric Oxide (NO) and Endotbelinl (ET1 ) in the circulation and brain tissue in models of acute brain ischemic and sham-operated rats, and the levels of No and ET1 in the circulation arid brain tissue in acute brain ischemic models with nimodipine. It was found that in compared with Sham -operated group, the levels of No in circulation of the model group had a tendency to decrease, but had no statistical difference (P>0. 05). the levels of ET1 was significantly increase (p<0. 05) ; in brain tissue, the levels of NO and ET1 were all inereased. Nimodipine has definite effects on the increasing the levels of NO in circulatin of the models. decreasing the levels of ET1, inhibiting the increase of NO and ET1 in brain tissue, It was suggested that nimodipine may improve thebrain blood flow, prevent the bain damage resulting from the acute brain ischemia.

尼莫地平是一种钙离子拮抗剂,现巳用于急性脑缺血的治疗。本文通过观察大鼠急性脑缺血模型及假手术组循环中及脑组织内一氧化氮(NO)、内皮素(ET1)水平,并观察尼莫地平对大鼠脑缺血模型循环中及脑组织的NO、ET1水平的影响,发现模型组循环中的NO水平较假手术组有下降趋势,但无明显统计学差异(P>0.05),ET1的水平较假手术组显著升高(P>0.05);脑组织中NO、ET1含量均较假手术组显著升高(P<0.05)。尼莫地平具有显著升高急性脑缺血大鼠循环中NO,显著降低循环中ET1,显著降低脑组织中NO、ET1的水平,说明尼莫地平具有促进脑缺血时大鼠的脑部供血,保护由于脑缺血引起的脑组织损害。

Objective To study the cerebral protective effect mechanism of naloxone in acute brain ischemic injury. Methods The rat focal cerebral ischemia models of middle cerebral artery (MCA) were established with the suture method. Using the technique of reverse transcription polymerase chain reaction (RT PCR),we detected the changes of Interleukin 1β messenger ribonucleic acid (IL 1β mRNA) level in brain tissue homogenate. Results The levels of brain tissue homogenate IL 1β mRNA in Saline treated group (ST) and Naloxone treated group (NT) were all significantly higher than that of Sham operated control group (SO) ( P < 0.01 ). Compared with ST group, the levels of IL 1β mRNA in NT group were significantly lower ( P < 0.05 ). Conclusion Administration of naloxone, an opioide receptor antagonist, has the brain protective effects in acute cerebral ischemia, by decreasing the level of IL 1β mRNA and reducing the acute cerebral ischemia injury caused by IL 1β.

目的 研究阿片受体拮抗剂纳络酮 (Naloxone)对急性脑缺血损伤的保护作用机制。方法 线栓法制作Wistar大鼠局灶性大脑中动脉缺血 (MCAO)模型 ,应用逆转录聚合酶链反应技术测定各组脑组织匀浆白细胞介素 1β信使RNA(IL 1βmRNA)水平的变化。 结果 生理盐水治疗组IL 1βmRNA水平显著高于假手术组 (P <0 .0 1) ;纳络酮治疗组IL 1βmRNA水平显著低于生理盐水组 (P <0 .0 5 )。结论 阿片受体拮抗剂纳络酮可通过降低脑内IL 1βmRNA水平来减轻IL 1所致的神经功能损害 ,从而起到脑保护作用。
