Objective: To investigate photoreceptor cell (PRC) apoptosis and gene regulation in hereditary retinal degeneration of Royal College of Surgeon (RCS) rats. Methods: The retinal sections of RCS rats in different ages were examined by light microscope, TdTmediated biotindUTP nickend labeling(TUNEL) and p53 protein immunohistochemical method. Results: Compared with age matched SD rats, PRC in RCS rats after being born for 15 days, gradually had outer segment membrane accumulation, inner segment disorder, nuclei pycnosis, and cell disappearance. Postnatal 30 to 40 days is the peak stage of PRC death. From 25 to 40 days, the nuclei of PRC in RCS rats were stained positively by TUNEL and the retina in the 35th day had the largest amount of stained nuclei. Positive staining of p53 protein could be seen in the retinal PRC of RCS rats at 25 to 35 days and showed the most at 30 days. The stained granules of p53 protein mainly deposited on the inner segment and some on the nuclei of PRC. Conclusion: It is through apoptosis that the retinal PRC died in retinal degeneration of RCS rats. Apoptosis of PRC in RCS rats might be p53dependent.

目的:研究遗传性视网膜变性中感光细胞(photoreceptorcel,PRC)凋亡及其基因调控。方法:对出生后9、15、20、25、30、35、40、60、80、100dRCS大鼠(RoyalColegeofSurgeon)及同龄SD(SpragueDawley)大鼠各4只的视网膜组织结构进行光镜观察、TUNEL(TdTmediatedbiotindUTPnickendlabeling)检测和p53蛋白免疫组织化学检测。结果:与同龄SD大鼠相比,RCS大鼠从出生后15d,PRC相继出现外节膜盘堆积、内节排列紊乱、细胞核固缩、细胞消失。30~40d是PRC消失的高峰期;出生后25~40d,有PRC细胞核呈TUNEL阳性染色,35d时,阳性数最多;出生后25~35d,PRC层(主要为PRC内节)呈p53蛋白免疫组织化学染色阳性,30d时染色最强。结论:RCS大鼠视网膜变性过程中,PRC以凋亡的形式死亡,并可能为p53依赖性。

To determine the process of photoreceptor cells (PRC) degeneration in hereditary retinal degeneration, the retinas of RCS rats and Sprague Dawley (SD) rats were examined by light microscope and TdT mediated biotin dUTP nick end labeling (TUNEL). Compared with age matched SD rats, the retinal PRC of RCS rats began to degenerate from 15 days of age. The debris of the outer segment disk membrane were accumulated on the surface of the retinal pigment epithelial cell; the inner segment were shortened and disordered; the nuclei underwent pyknosis. From 30 to 60 postnatal days, PRC disappeared quickly and only few were remained. TUNEL study showed that the positive labeled nuclei of PRC progressively increased from 25 days of age, and got the maximum level at 35 days of age. In inherited retinal degeneration of RCS rats, PRC progressively lose the normal structure and finally die through apoptosis. The peak phase of degeneration of PRC is from postnatal days 30 to 40.
