Study the method of quantitative/identified detection of genetically modified (GM) maize Mon810 components infoods and in feeds by real-time PCR. 采用实时荧光PCR技术,建立了定量(性)鉴定检测加工产品中转基因玉米Mon810成分的方法。实验设计的可以扩增玉米自身基因和外源基因边界序列的引物和探针具有品种和品系特异性,特异性地检测出食品、饲料等加工产品中转基因玉米Mon810成分。某些检测样品不仅检出转基因玉米Mon810成分,还同时检出其它转基因玉米品系或其它转基因品种。本研究实验建立的转基因玉米Mon810品系鉴定检测方法,即可以用于加工产品中转基因成分的定量检测(检测低限为0.1%),也可以用于定性检测,或作为常规PCR定性检测后的确证实验方法。

本研究成功建立了商业化种植的转基因玉米MON810的筛选检测和品种鉴定的PCR方法。该方法根据玉米自身IVR基因作为内源特异参照基因扩增226bp片段,检查模板DNA提取的质量,避免了假阴性结果;同时扩增CaMV35S启动子基因195bp片段,可以对MON810转基因玉米进行筛选检测;此外,根据MON810品种设计的特异性鉴定检测引物,可扩增Maizegenome/CaMV35S基因170bp片段和HSP/CryIA(b)基因194bp片段,具有很强的品种特异性,达到了对MON810转基因玉米的品种鉴定目的。PCR反应循环参数是94℃2min;94℃40sec,55℃60sec,72℃60sec,35次循环;之后72℃延伸5min。
