Objective To observe the changes of the rates of the CD4 +、CD8 + cell which express cytokine IFNγ and IL 4 in systemic lupus erthematosus(SLE). Method Lymphocyte was stimulated and cultured while adding protein transit inhibitor, then fixed by fluorescence labled monoclonal antibody to the antigen on cell surface and anti cytokine monoclonal antibody. With the fixing and broking membrance technology, the CD4 +、CD8 + cell which yield IFNγ and IL 4 were detected. Results We detected the rates of the CD4 +、CD8 + cell which express cytokine IFNγ and IL 4 in lymphocytes of 60 healthy volunteers and 30 cases with SLE. The reference range of our labortory was obtained and we found the rates were obviously abnormal in SLE. Conclusion IFNγ CD4 +、IFNγ CD8 + cell have important significance in the course of SLE.

目的 观察系统性红斑狼疮 (SLE)患者表达IFNγ及IL 4的CD4 + 、CD8+ 细胞的改变。方法 选SLE患者 30例和健康成人 6 0例 ,用刺激剂激活淋巴细胞同时加入蛋白转运抑制剂进行细胞培养 ,再用荧光标记的抗细胞表面抗原单抗及抗细胞因子单抗结合固定、破膜处理技术及流式细胞术检测产生IFNγ及IL 4的CD4 + 、CD8+ 细胞 ,同时以相应同型IgG染色细胞作为阴性对照以确定细胞因子染色的特性。 结果 SLE患者表达IFNγ的CD4 + 、CD8+ 细胞百分率分别为 (6 .92± 2 .84 ) %和 (7.0 9± 2 .5 7) % ,与健康成人 [(3.88± 1.13) %、(4.5 1± 2 .17) % ]相比 ,显著升高。表达IL 4的CD4 + 、CD8+ 细胞百分率在健康成人组与患者组间差异无显著性。结论 表达IFNγ的CD4 + 、CD8+ 细胞在SLE的发病过程中具有一定的意义
