Bone marrow cells collected from normal donors and patients with chronic myelogenous leukemia (CML) and leukemic cell line (K562) have been investigated after the treatment with different concentration of hematin. The effect on the growth inhibition of clonogenic cells was observed in bone marrow cultures and leukemic cell line(K562), with the combined effect of Hematin and γ radiotion. The details of the principle of Mossbauer inactivation, experimental method, and experimental resrlts are given.

To assess the potential of photochemotherapy for the in vitro purging of residual leukemia cells from autologous bone marrow transplants ,we studied the effect of YHPD combined with light irradiation on human leukemia cell line K 562 and normal human hematopoietic progenitor cells by using the methods of trypan blue exclusion test and colony assay. We also investigated the influence of YHPD dose and light exposed time on this photosensitivity of K 562 cells. It was found that YHPD or light irradiation alone had no apparent cytotoxicity while the application of YHPD plus light exposure showed the photosensitization. Decrease in cell viability was linear when plotted on a logarithmic scale from 0 to 5 min (light irradiation) or 0 to 5 mg/L(YHPD). YHPD plus light irradiation was selectively more efficient for killing K 562 cells than normal human marrow progenitors. Under the condition of a 5log reduction in the survival of K 562 cells, 15.97% of normal human CFU GM survived. These results indicate that the photochemotherapy mediated by YHPD might be used for purging bone marrow in vitro.

为研究光卟啉(YHPD)加光照对白血病细胞株K562的杀伤作用及对正常造血祖细胞的影响,通过细胞体外液体培养活细胞计数、体外半固体培养集落测定,以探讨该法用于体外净化自体移植骨髓中残留的白血病细胞的可能性。观察不同光敏剂剂量和光照时间的变化对杀伤白血病细胞作用的影响,结果表明,单用YHPD或单独日光荧光照射对K562细胞和正常人造血祖细胞均无明显杀伤作用。二者联合使用表现光敏效应。细胞存活率的对数与YHPD剂量、光照时间呈线性相关。YHPD加光照对K562细胞具有强大杀伤作用,对正常造血祖细胞影响较小,在能够杀伤白血病细胞大于5个对数级,其体外已不能形成集落的情况下,正常人骨髓-巨噬细胞集落形成单位(CFU-GM)尚有15.97%,提示该法可望用于骨髓体外净化

Methods The MSC were isolated from the nucleated cells fraction of autologous bone marrow by density gradient centrifuge, and then induced into chondrogenic differentiation by adding dexamethasone, transforming growth factor β 1(TGF β 1) and ascorbic acid in vitro . After 3 weeks, some cells turned to round shape and secreted metachromatic matrix. The cartilaginoid grafts composed of chondrogenic MSC. Bovine type Ⅰ collagen and human fibrin were cultured within the chondrogenic medium for 2 weeks in vitro or transplanted subcutaneously adjacent to the knee joint for 3 weeks in vivo . Results The most cells in the grafts were degenerated and disappeared after cultured in vitro . But the residual cells were survival and secreted metachromatic staining proteoglycan with toluidine blue, which was characteristic cartilage matrix. The grafts developed into matured cartilage tissue assessed by histological examination after 3 weeks of transplantation in vivo . Conclusion MSC can be used as functional cells to constructing tissue engineered cartilage.

目的 采用组织工程方法 ,以培养后的兔骨髓间质干细胞 (MSC)制成人工软骨培养物 ,经体内外培养后发育出成活的软骨组织。方法 抽取兔骨髓液经密度梯度离心得到单个核细胞 ,再经体外分离、培养获得兔骨髓 MSC。向 MSC培养液内加入地塞米松、转化生长因子 -β1 (TGF-β1 )和维生素 C进行软骨起源诱导培养 3周 ,部分细胞开始转变为圆形并分泌基质。将诱导后的细胞与牛 型胶原及人纤维蛋白按一定的比例混合 ,制成软骨样的培养物并分别做体内外培养。结果 体外培养 2周后 ,培养物内大部分细胞已萎缩消失。但剩余的少量细胞成活 ,形成类似的软骨陷窝并分泌甲苯胺蓝异染的软骨基质。体内移植培养 3周后 ,培养物已发育成颗粒状成熟的软骨组织。结论 骨髓间质干细胞可用于组织工程软骨组织的构建 ,是一种非常有前途的人工软骨组织构建中的功能细胞。
