Objective To investigate the way of interstitial cell loss and its possible relation to Fas expression during the course of renal interstitial fibrosis. Methods The model was induced by unilateral ureteral ligation in rats and the rats were sacrificed at postoperative days 3, 7, 14, 28, 56, and 84. Besides routine morphologic examinations, interstitial collagen type Ⅲ and Fas expression was detected by immunohistochemistry staining and apoptotic cells were determined by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL). Results Both of the Fas expression and the apoptotic cells in the renal interstitium were continuously increased from day 14 to the end of experiment, with a parallel increase of the interstitial fibrosis. There was a positive correlation between the Fas expression and apoptotic cells in renal interstitium( r=0 94, P<0 05). Conclusion The apoptosis in this model may be pathogenetically related to interstitial cell loss in the course of renal interstitial fibrosis and the apoptosis is probably mediated by Fas/FasL system.

目的探讨细胞间粘附分子1(ICAM1)在肾小管间质炎性病变中的作用。方法将荧光标记的ICAM1反义寡核苷酸经静脉注射至正常小鼠体内,观察其分布状况。小鼠在行单侧输尿管梗阻术后从尾静脉注射ICAM1反义寡核苷酸(7只),术后第7天留取输尿管梗阻侧肾脏检测肾组织ICAM1mRNA的表达并作病理检查。同时设立对照寡核苷酸(8只)和注射用水组(6只)作为空白对照。结果ICAM1反义寡核苷酸经静脉注射后主要分布于小鼠的近端肾小管上。与对照组小鼠比较,经反义寡核苷酸治疗的小鼠梗阻肾组织内ICAM1及Mac1阳性细胞的表达明显减少,肾组织中ICAM1mRNA的表达也明显减少,肾小管间质中炎细胞浸润数比对照组明显减轻(110±14个/mm2,270±13个/mm2)、间质增宽的相对面积[(75%±09)%、(254±40)%]、基质积聚及纤维化等病变均明显减轻。治疗组小鼠在肝脏、脾脏重量、血尿素氮、肌酐、丙氨酸转氨酶及尿蛋白的检测上与对照组小鼠相比较差异均无显著意义。结论在非免疫因素引起的肾小管间质损伤中,ICAM1所介导的细胞粘附反应具有重要的作用,ICAM1反义寡核苷酸对小鼠单侧输尿管梗阻?

Objective To determine the relationship between P-selectin and unilateral ureteral obstruction (UUO). Methods The effect of anti-P-selectin monoclonal antibody (P-selectin mAb) on the renal histopathological changes and P-selectin expression was investigated in the UUO experimental model. Results Renal interstitial macrophage infiltration was observed within 24h after UUO, with marked increase at day 3 and a peak response at day 7. Interstitial edema was also found at day 7. The expressin of P-selectin was upregulated on the tubular epithelium and within the interstitium as early as 24h after ureteral ligation, with a peak expression at day 3 and down-regulated at day 7. Macrophage accumulation was significantly inhibited when P-selectin was blocked by mAb. Conclusion P-selectin plays a role in interstitial macrophage infiltration in the early phase of UUO, and P-selectin mAb might improve the interstitial leisons.

目的探讨粘附分子P -选择素在单侧输尿管梗阻后肾炎中的作用。 方法建立单侧输尿管梗阻(UUO)动物模型 ,观察肾组织病理改变和P -选择素在肾组织中的表达 ,以及抗P -选择素单克隆抗体 (单抗 )对其影响。 结果大鼠UUO第 1d即见间质以单核巨噬细胞为主的炎细胞浸润和P -选择素在小管间质表达 ;第 3d间质炎细胞增多 ,P -选择素明显表达 ;第 7d间质炎细胞浸润达高峰伴间质水肿 ,而P -选择素表达下降。抗P -选择素单抗治疗组炎细胞浸润减轻 ,P -选择素表达减弱。 结论P -选择素在UUO早期参与介导以单核巨噬细胞为主的炎细胞在肾间质浸润及肾间质损害 ,抗P -选择素单抗对此具有一定治疗作用。
