The composition of the semen dilutor was 8% glucose solution 77 ml, glycerin 3 ml and yolk 20ml, The diluted semen went into the prefreezing-equilibration and semen temperature fell from 20-30℃(room temperature)to 10-15℃ in 3-4 hours, then continually fell to 7-9℃ in 1-2 minutes. The thawing solution was mainly composed of 5% glucose solution 100ml and 10% Caffeinum Natrio Benzoicum 5ml. When frozen-thawed semen dosage of 15-20ml containing 1-1.5×10~9 motile spermatozoa-was deposited by single or double insemination in every oestruso farrowing rate of 682 sows was 73.75%. The mean litter size of 503 farrowed sows was 10.50. 7 sows were inseminated with the frozen semen stored for 3 years and all were pregnant. Difference of fertility rate of frozen-thawed semen within 4 hours was not significant.

The boar semen samples collected from 9 Meishan,8 Erhualian and 4 SovietLarge White were cold-shocked and frozen-thawed to determine the effectsof cold shock and freezing-thawing on some properties of boar semen.Therewere 10 treatments in 4 groups,including raw semen(control),diluted,diluted andequilibrated and frozen-thawed semen.The results showed that the motility ofspermatozoa decreased markedly after cold shock,and became weaker and weakerwith the lapse of time.Under light microscope the morphological features of boar spermatozoaafter cold shock were distended plasma membrane,and elevated or wrinkledapical ridge.There were indications of protecting acrosomes from cold shock bydilution and equilibrium.Swelled,vesiculated and disrupted acrosomes were foundafter freezing-thawing.The positive correlation was found significant betweenthe percentage of normal acrosome and the motility of spermatozoa in the eja-culates(r=0.60,p<0.01),while no significant relationship between the two wasfound after lowtemperature treatments.The ultrastructural investigation showed that the damaged region of boarspermatozoa after freezing-thawing appeared to be on the acosomes.Severelydistended or disrupted plasma membrane,swelled and vesiculated outer acosomalmembrane and dispersion of the acosomal content were the prominent featuresof damaged boar spermatozoa after freezing-thawing.No so called“hybridvesicles”fused by plasma membrane and outer acosomal membrane were found inthe investigation.The GOT and LDH activities in seminal plasma increased after cold shockand freezing-thawing.After frozen-thawed treatments LDH was 1.5 times,andGOT was 16.3 times as many as in raw semen.No apparent changes were foundin GPT activities after low-temperature treatments.Five clear isoenzymaticbands were separated in boar seminal plasma by means of agarose gel electro-phoresis.There were some changes in the LDH isoenzymatic patterns,such asthe increased percentage of LDH-4 in raw seminal plasma after cold shock(p<=0.05),the same change in the frozen-thawed seminal plasma(p<0.01),andthe increased M-LDH after dilution and equilibrium.
