Measurement of bone alkaline phosphatase and relative study with osteosarcoma
      
The objective of this paper is to explore the value of bone alkaline phosphatase (BALP) for diagnosing osteosarcoma, evaluating the effect of the chemotherapy, judging the prognosis and supervising the relapse and metastasis.
      
The immunoassay was used to check the BALP of the blood serum that was from 42 primary osteosarcoma patients.
      
BALP was more sensitive than ALP in diagnosing osteosarcoma (P = 0.015).
      
It has applied value in the diagnosis of osteosarcoma, reflection of the effect of chemotherapy and forecast the prognosis.

Scanning electron microscopy studies showed that the cells were irregular in shape and the cell surface was covered with large number of microvilli.Sometimes,a few of ziotic blebs were observed.It seemed that the cells are attached to the glass by lameli- podia and filopodia of variable lengths. Transmision electron microscopy studies showed that the cells had large nuclei with prominent nucleoli.Their cytoplasm contained abundant free ribosomes,prominent Golgi complexes,a few mitochondia and scattered dense lysosome-like bodies.The rou- gh endoplasmic reticulum was often expanded into many vacuoles,filled with modera- tely dense amorphous meterials.No calcuium apatite deposits and virus particles were observed. The ultrastractural localization of phosphatase activity studies demenstrated that most of the cells showed deposits of reaction products of alkaline phosphatase on the whole circumference of reaction products of alkaline phosphatase on the whole circum- ference of sectioned plasma membrane or as scattered foci.Sometimes,deposits of rea- ction products were also observed on the nuclear membrane.No deposition of reaction products of acid phosphatase was observed except within some lysosomes.

Ten cases of human osteosarcoma have been studied by light microscopy, electron micros-copy and energy dispersive X-ray spectrometer (EDX). Ten cases of human osteosarcoma have been studied by light microscopy, electron micros-copy and energy dispersive X-ray spectrometer (EDX). Ultrastructurally, osteosarcoma cells areclassified into five types: osteoblastic type. chondroblastic type, fibroblastic type, primitivemesenchymal cell and multinucleated giant cell. All the adovetumor cells are originated fromthe multipotential primitiv mesenchymal cell. The characteristic feature of malignant cells: besides common morphologic characteristics,there are often the intranuclear tube-like inclusion bodies and many electron-dense granulesand directionless microfilaments in the cytoplasm. On the intramatrical collagen fibers are depo-sited many hydroxyapatite crystal particles. EDX analysis showed the elemental compositions of ossiform tissue exist in the tumortissue. The formation of hydroxyapatite crystal particles indicated the initial stage of local cal-cification.

对10例人骨肉瘤进行了光镜病理学,电镜超微结构和能谱元素成份的研究。在超微结构上,将骨肉瘤细胞分类为骨母细胞型,软骨母细胞型,纤维母细胞型,原始间叶细胞和多核巨细胞等五型。各型瘤细胞的组织发生均来源于多能性原始间叶细胞。瘤细胞的恶性特点,除具有肿瘤性共同的形态特征之外,核内常见管状包涵体,胞质里有较多散在致密颗粒和无定向排列的微丝,间质胶原纤维中沉积着羟基磷灰石结晶颗粒。能谱分析说明瘤组织内有骨样组织的元素成份,羟基磷灰石结晶颗粒的形成反应了初期局部钙化。

Through fusion between the splenic cells from BALB/C mice (after primarily in-trasplenic immunized with HOS-8603 human osteogenic sarcoma cells) and mouse myeloma cells NSO, then applying ELISA and immunoflourescence staining for screening, we succeeded in the establishment of two hybridoma cell lines capable of secreting anti-osteosarcoma monoclonal antibodies. These two cell lines were named OS-MAb, and OS-MAb2. This report is mainly on the OS-MAb1 cell line. Immunohistochemical study showed that the McAb secreted by OS-MAb1 also reacted positively with a certain proportion of giant cell tumors of bone, chondrosarcomas and Ewing's sarcomas in addition to osteoarcomas, but negatively with other tested tumors as well as normal adult and fetal tissues. Monoclonal antibody from OS-MAbj labelled with 125I was injected into athymice nude mice with xenografts of human osteogenic sarcoma. Three to four days after injection,the osteogenic sarcoma xenografts could be clearly visualized by Eadio-Immuno-Imaging. The present study suggests that the antibody from OS-MAb, can be of clinical value for the detection or treatment of osteogenic sarcoma.

用人体骨肉瘤细胞株HOS-8603脾内一次性免疫BALB/c小鼠,取其脾细胞与骨髓瘤细胞 NSO融合,经ELISA和 FIA筛选,获得2株分泌抗人体骨肉瘤单克隆抗体(McAb)的杂交瘤细胞株(OS-MAb_1)和OS-MAb_2)。用ABC 免疫酶标法检测发现(OS-MAb_1)杂交瘤细胞所分泌的 McAb,除对人体骨肉瘤组织呈阳性反应外,对部分骨巨细胞瘤、软骨肉瘤和尤文氏肉瘤组织呈阳性反应,而对其它肿瘤组织和正常成人及胎儿组织呈阴性反应。同位素~(125)I标记该抗体,在荷人骨肉瘤裸鼠体内定位,发现72小时起到96小时肿瘤放射性影像清晰,放射性积聚高于其他部位。显示OS-MAb_1株所分泌的抗人体骨肉瘤McAb作为定位诊断和导向治疗载体的可能性。
