    A series of hybridoma cell lines producing monoclonal antibodies to DNA had been obtained by hybridization of spleen cells from MRL-1Pr/1pr mice, which had developed a severe form of SLE and spontaneously produced high levels of antibodies to DNA without exogenus immunization, with SP_2/0-Ag-14 plasmacytomas. The McAb H241, one of those monoclonal antibodies, was used to immunize NZW rabbit and an anti-H241 idiotypic antibody was harvested from immune serum of the rabbit successfully. The sensitivity of H241 and idiotypic cross-reactivity among H241 and other 24 monoclonal anti-DNA autoantibodies in reaction with H241 and anti-H241 idiotypic sera were investigated by radioimmunoassay (RIA). The results demonstrated that H211 had a high sensitivity, 6ng of unlabled H241 was required to achieve 50% inhibition of binding of labeled H241 to anti-H241 idiotypic serum. The experiments also demonstrated that 15 of 24 other monoclonal anti-DNA antibodies represented idiotypic cross-reactivity with H241 in different degrees. These results suggest that although frequently idiotypes have diversity, some antibodies on the basis with similar structure of the idiotypic determinants were limited in the extent of diversity. It is possible that the lupus mouse embryo genes controlling immunoglobuling synthesis involve correlatative loci for some anti-DNA autoantibodies.

Antibody against HSP4 3 and autoantibodies against double strained DNA (dsDNA),single strained DNA(ss-DNA),ENA,and histons were detected by ELISA method.Had found that serological changes in Balb/c mice immunized with HSP43 were similar to the counterparts in the SLE-like animal model induced by chronic infection or immunization with CJ-S131 and in the spontaneous lupus mice,summarized as ① autoantibody against dsDNA started to go up in the 4th week,and peaked in the 6th week,the second peak in the 16th week;② autoantibodies against ss-DNA and ENA went up gradually,the peak responses were achieved in the 16th week;③ peaked in 8th week,the kinetic of autoantibody against histons was different from those of above three autoantibodis;④positive percentages of mice with elevated autoantibodies against ds-DNA,ss-DNA and ENA were higher than that of autoantibody against histons in the 4th,6th,16th week,but not in the 8th week;⑤no autoantibody against RNA was detected;⑥isotypes of autoantibodies were IgG and IgM;⑦the rising of those autoantibodies was the result of injection of HSP43 and relevant to the dosage of injection. The results suggested that specific antigen might not be needed after autoreactive lymphatic clones were selected and activated to some degree,This study provided an experimental evidence for the hypothesis that immune response against HSP may be highly cross-reactive and even involve autoimmune diseases.

    小鼠每周注射1次空弯菌HSP43共10w。HSP43抗体第4周起显著升高─直持续到16w,抗ds-DNA、ss-DNA自身抗体在6w和16w出现两个高峰,抗DNA自身抗体在4~16w逐渐升高,抗组蛋白抗体在8w最高然后下降,单个小鼠自身抗体反应格局呈现4种类型。各类自身抗体的出现和升高与注射HSP43剂量有依赖关系。该结果为自身免疫病的热休克蛋白发病假设提供了直接的实验证据。

    Aim To investigate whether there is IL-12 signaling pathway through lck/p38/c -jun in splenic cells obtained from lupus mouse and its effect on splenic cells. Methods Mice with graft versus host disease were used as lupus nephritis model. Activity of Lck tyrosine kinase, p38 phosphorylation and mRNA expression of c-jun in splenic cells were determined by autoradiography, Western blot and Northern blot, respectively. Results There were higher levels of Lck activity, p38 phosphorylation and c-jun expression of IL-12-stimulated splenic cells from lupus model when compared with that observed in similarly treated splenic cells from normal control. The Lck activity and p38 phosphorylation were almost inhibited by Lck inhibitor PP1, on the other hand, p38 specific inhibitor SB203580 decreased phosphorylation of p38. In addition, expression of c-Jun was also inhibited by PP1 or SB203580 although splenic cells were stimulated with IL-12. Conclusion Aberrant murine splenic call, IL-12-me- diated intracelluar signaling pathway through lck/p38/c-Jun were involved in immunologic damage.

    目的探讨在狼疮小鼠脾细胞中是否存在IL-12通过Lck/P38/c-jun传递信号及其对脾细胞的影响。方法以慢性移植物抗宿主病(graft versus host disease, GVHD)小鼠为狼疮小鼠模型,分别采用放射自显影、 Western blot和Northern blot,检测培养的脾细胞中 Lck的活性、 p38磷酸化活化及c-jun基因表达的变化。结果狼疮小鼠脾细胞经IL-12刺激后,与正常对照组相比较,Lck的活性增高、p38异常活化,c-jun mRNA的水平增高。加入Lck抑制剂 PP1组Lck的活性及p38磷酸化消失,无c-jun基因的表达;加入p38抑制剂SB203580组亦无p38磷酸化及c-jun基因的表达。结论狼疮小鼠的脾细胞异常,IL-12可通过Lck/p38/c-jun在细胞内传递信号,直接参与免疫损伤。
