Aim To investigate whether there is IL-12 signaling pathway through lck/p38/c -jun in splenic cells obtained from lupus mouse and its effect on splenic cells. Methods Mice with graft versus host disease were used as lupus nephritis model. Activity of Lck tyrosine kinase, p38 phosphorylation and mRNA expression of c-jun in splenic cells were determined by autoradiography, Western blot and Northern blot, respectively. Results There were higher levels of Lck activity, p38 phosphorylation and c-jun expression of IL-12-stimulated splenic cells from lupus model when compared with that observed in similarly treated splenic cells from normal control. The Lck activity and p38 phosphorylation were almost inhibited by Lck inhibitor PP1, on the other hand, p38 specific inhibitor SB203580 decreased phosphorylation of p38. In addition, expression of c-Jun was also inhibited by PP1 or SB203580 although splenic cells were stimulated with IL-12. Conclusion Aberrant murine splenic call, IL-12-me- diated intracelluar signaling pathway through lck/p38/c-Jun were involved in immunologic damage.

目的探讨在狼疮小鼠脾细胞中是否存在IL-12通过Lck/P38/c-jun传递信号及其对脾细胞的影响。方法以慢性移植物抗宿主病(graft versus host disease, GVHD)小鼠为狼疮小鼠模型,分别采用放射自显影、 Western blot和Northern blot,检测培养的脾细胞中 Lck的活性、 p38磷酸化活化及c-jun基因表达的变化。结果狼疮小鼠脾细胞经IL-12刺激后,与正常对照组相比较,Lck的活性增高、p38异常活化,c-jun mRNA的水平增高。加入Lck抑制剂 PP1组Lck的活性及p38磷酸化消失,无c-jun基因的表达;加入p38抑制剂SB203580组亦无p38磷酸化及c-jun基因的表达。结论狼疮小鼠的脾细胞异常,IL-12可通过Lck/p38/c-jun在细胞内传递信号,直接参与免疫损伤。

Peripheral blood cells were observed, CD 3 -CD 5 + and CD 3 +CD 5 + of recipients were measured by flowcytometry. Recipients' mixed chimerism were analyzed with polymerase chain reaction short tandem repeat and recipients'kindey pathological changes were examined with direct immunofluorescence after transplantation. The lowest values of WBC, PLT and Hb fall in the NST group were (0 6±0 2)×10 9/L, (27 0±7 6)×10 9/L and (48 0±1 2) g/L, respectively. They were all higher than those of the control group(P all < 0 05).The time for WBC rising from the lowest value to 1 0×10 9/L were 17 5±6 3 days in the NST group, which was shorter than that in the control group(P<0 05). There were no statistically significant difference in the reduction of urinary excretion of proteins, change of anti d sDNA and reduction of kindey immunofluorescence between the two groups(P all >0 05). Early immune reconstitution began to recover 8 weeks after transplantion in the NST group. The incidences of bleeding, loss of body weight, penumonia and mortality related to transplantation were lower in the NST group than those in the control group (P all<0 05). Recipients' mixed chimerism in the NST group appeared 30 days after transplantation. [Conclusion] NST can improve all manifestations of lupus mice effectively. NST show less suppression to bone marrow function. Hematopoietic function recovers faster with NST than with Allo SCT. Incidences of complications and mortality related to transplantation in the NST group are lower than that in the Allo SCT group. NST is a safe and effective method for the treatment of lupus mice.

Objective To detect the expression of "regulated upon activation normal T cell expressed and secreted" (RANTES) in kidney tissue of murine chronic graft versus host disease (GVHD) lupus nephritis,and investigate the prednisone modulation on RANTES.Methods The protein and mRNA expressions of RANTES were detected by immunohistochemistry and in situ hybridization,respectively.Results The protein and mRNA expressions of RANTES were significantly increased in kidney tissue of GVHD mice,respectively,which could be dectected in glomerular,tubular and interstitial cells,especially much in tubular epithelial cells and interstitial mononuclear cells.The protein expression of RANTES in kidney tissue was correlated positively with its mRNA expression ( r =0 612, P <0 05).Prednisone treatment significantly prevented the increase in urinary protein excretion,anti dsDNA antibody,glomerular cell proliferation,and interstitial cell infiltration in the lupus mice,indicating the beneficial effects of prednisone on this lupus nephritis.Prednisone significantly suppressed the increased renal RANTES expression (protein and mRNA).Conclusion The results provide the evidence of the marked increase in renal RANTES expression in murine GVHD lupus nephritis.The beneficial effect of prednisone on this lupus nephritis may be partially mediated by the suppression of RANTES.
