采用改良的CTAB方法提取转基因甘蔗幼苗的基因组DNA ,核酸蛋白测定仪分析及凝胶电泳结果表明 ,提取的DNA具有典型的DNA分子的标准紫外吸收光谱特点 ,DNA产量为 4 5~ 6 0 μg/ 10 0mg。同时根据质粒载体设计合成了 2对引物 ,分别扩增新霉素磷酸转移酶 (nptII)基因和甘蔗花叶病毒外壳蛋白(ScMV CP)基因。PCR检测结果表明 ,在 5 3株待检测的样品中有 2 4株同时含有这 2种转基因成分 ,其中 13株在Southern杂交中有杂交信号出现 ,其拷贝数为 1~ 3个。
