Four single nucleotide polymorphisms(T423C,A501G,C511A,A610G) were found, and the C511A and A610G nucleotide substitution resulted in 2 amino acid changes(Lysine171Glutamine and Alanine204T hreoine).
Multiple shoots were induced on differ- entiation medium of MS supplemented with IAA 0.1mg/L,ZT 2.0 mg/L,adenine 40 mg/L and glutamine 100 mg/L,and the differentiating rate reached 20%.
Exogenous glutamine and proline accumulated over 2 h at 4°C in swelling seeds were spent at different rates over the following 2 h at 20°C, thus compensating for insufficiency of these amino acids during the initial stage of development.
Creation of additional pools of glutamine and proline during the initial stage of amino acid metabolism had positive effects on the seed germination and vital activity of the plants.
The glutaminase (EC 3.5.1.2) isolated from seedlings of triticale (Triticalesp.) had a pH optimum of about 8, was inhibited with excess substrate (glutamine), and reaction products (glutamate and NH+4).
(1) In order to avoid the postmortem changes Richter and Dawson and proposed to fix the brain in situ with liquid air. We found that a mixture of solid carbon dioxide and acetone was a serviceable substitute for liquid air, at least for the purpose of ammonia analysis. (2) A modification of Conway's micro-diffusion method for the determination of ammonia was described. The modified method enabled us to determine ammonia in the range from 0.05 to 0.50μg. (3) Conway's "half-diffusion" method was adapted to mi...
(1) An effective and rapid method for the separation of ammo acids by a combination of high-voltage electrophoresis and high-temperature chromatography is described. The conditions for electrophoresis were: formic-acetic acid buffer, pH 1.9, 0° to-2℃, 33V/cm, 2 hours. The solvent system used for high temperature paper chromatography was n-BuOH: pyridine: 60% EtOH:H_2O=5:1:1:1.The full course took ten hours only. Satisfactory resolution was obtained with serine and glycine, and with histidine, arginine and l...
It was found in our previous work that thymo-lymphosarcoma exhibited a much higher glutaminase activity than the normal thymus. In this investigation, a comparative study has been made on the kinetic properties of glutaminase in the mitochondria of normal thymus and thymo-lymposarcoma. No significant differences were observed on pH optima, Michaelis constants and the responses to both inhibitors and activators, indicating that the enzymes may possess similar active centers. The enzymes from both normal and t...
