Objective To explore the mechanism of nasal tolerance by nasal administration of myelin basic protein (MBP) in guinea pigs for inducing immune tolerance and preventing experimental autoimmune encephalomyelitis (EAE). Methods Guinea pigs were, to begin with, given MBP through the nasal route to induce the development of peripheral immune tolerance, and then immunized with specific antigen MBP plus complete Freund's adjuvant (CFA). The clinical symptoms of guinea pigs were investigated, and the erythrocyte rosette test, and lymphocyte blastogenesis test, the MBP specific skin delayed type hypersensitivity (DTH) response and the levels of interlukin 4 (IL 4) and interferon γ (IFN γ) were also detected, and the pathological changes were examined as well. Results Nasal tolerance could effectively prevent the development of EAE clinically. The erythrocyte rosette test, lymphocyte blastogenesis test and MBP specific skin DTH response were also obviously suppressed. An evident increasing of IL 4 level while a decreasing of IFN γ level was found after the culture of peripheral blood lymphocytes. Conclusion This experimental study suggested that the nasal administration of small amount of antigens had been a highly efficient approach in inducing clinical relevant tolerance, and it might provide a simple and promising way for the protection and treatment of a number of many autoimmune diseases, including EAE. It was suspected that the suppression of EAE in this experimental study might be associated with down regulation of the Th1 response or secretion of proinflammatory cytokines, i.e., IFN γ and with up regulation of the Th2 response or secretion of anti inflammatory cytokines, i.e., IL 4.

目的研究激活诱导细胞死亡 (AICD)现象在乙型肝炎慢性化和重型化机制中的意义。方法分离 2 0例慢性 /慢性重型乙型肝炎病人与 10例健康献血员外周血单个核细胞 (PBMC) ,在 PHA的刺激下培养 72 h后收集细胞 ,经流式细胞仪检测凋亡。结果 PBMC凋亡率乙型肝炎组明显高于正常对照组 [(2 5 .48± 14.0 7) % vs(11.45± 5 .2 7) % ,P<0 .0 1];慢性乙型肝炎组 [(30 .5 7± 13.43) % ]明显高于正常对照组 [(11.45± 5 .2 7) % ,P<0 .0 1]和慢性重型乙型肝炎组 [(13.5 9± 6 .44 ) % ,P<0 .0 1];PBMC凋亡率乙型肝炎 HBe Ag(+ )组明显高于正常对照组 [(2 9.5 0± 12 .5 4) % vs(11.45± 5 .2 7) % ,P<0 .0 1]。结论 AICD可能是形成 HBV慢性感染免疫耐受的一个重要机制。

目的研究乙型肝炎病毒(HBV)对外周血单个核淋巴细胞(PBMC)凋亡的作用。方法利用10名健康献血员外周血分离PBMC,分为加HBV阳性血清组和加健康人血清对照组,经培养72h后,采用PI染色法在流式细胞仪上检测细胞的凋亡情况。结果加HBV阳性血清组培养细胞的凋亡率为(39.56±7.03)%,明显高于加健康人血清组培养细胞的凋亡率(27.57±7.78)%,两组间具有非常显著的差异(P∨0.02)。结论HBV可能具有诱导PBMC凋亡的能力,这可能是形成HBV慢性感染免疫耐受的一个重要机制。
