Ribose was inactive and the free purine and pyrimidine bases were even slightly inhibitory.The increase in incorporation rate caused by a mixture of the four 2',3'-nucleotides was not greater than that produced by the most active one namely uridylic acid.Ribonucleic acid, either soluble or high molecular one, accelerated glycine incorporation. However, this effect was not observed consistently and varied with different batches of the spheroplast preparation.A comparatively minor fraction of the incorporated glycine was found in either the C-or the N-terminals of the proteins isolated from the spheroplasts, thus indicating that the amino acid was bound inside the protein molecules. The same result was noted when the incorporation rate was stimulated by the addition of 2',3'-uridylic acid.

Unlike the E. coli enzyme, the bovine pancreatic RNase can act without Mg~(++)and under such condition, SRNA can be degraded as fast as HRNA(sometimes even faster). However, when Mg~(++)is included in the incubation mixture, the rate of degradation of SRNA becomes much slower. On the other hand, HRNA is degraded even faster. This excludes the possibility that the slower degradation of SRNA is due to the inhibitory effect of Mg~(++)on pancreatic RNase. The protect effect of Mg~(++)on SRNA against degradation is observed even when the incubation temperature is raised to 60°C. Thus, we propose that the secondary structure of SRNA is responsible for its resistance to enzymic degradation and Mg~(++)has a profound stabilizing effect on its secondary structure.It has also been found that when RNA is added during precipitation with the uranyl acetate reagent, the acid soluble ultraviolet absorption becomes smaller. In other words, some of the acid soluble nucleotides are co-precipitated with RNA by this treatment. This may explain at least partially the observation that SRNA"inhibits"the enzymic degradation of HRNA both noticed by us and by Zillig.The stabilizing effect of Mg~(++)on the secondary structure of SRNA and in turn on the resistance of SRNA to enzymic degradation may have some bearing on the slow and incomplete degradation of SRNA by polynucleotide phosphorylase and snake venom phosphodiesterase since these enzymes all require Mg~(++)for activity.The physiological significance of the resistance to enzymic degradation of SRNA is briefly discussed.

A simple method is suggested for the preparation of relatively large quantity of soluble ribonucleic acid from E. coli by direct extraction of the bacterial cells with phenol, followed by fractionation with ammonium sulphate. The product is found to be homogeneous as judged by its electrophoretic and sedimentation patterns. It contains only adenosine as the terminal nucleoside and from the content of which, the chain is estimated to consist of 78 nucleotides. Its ability to accept S~(35)-methionine is...

Its ability to accept S~(35)-methionine is rather high as compared with that reported in the literature.The chemical and physico-chemical characteristics of the product, such as protein, polysaccharide, deoxyribonucleic acid and nucleo-base contents, specific rotation, ultraviolet absorption spectrum, and atom phosphorus extinction coefficient, have also been determined.
