After giving a brief account on recent advances of molecular ecology,this new field of ecological sciences has been characterized by using DNA markers.Based on molecular works on natural populations of animals and plants,which have been done in Chinese Academy of Sciences in recent 5 years,advances of molecular ecology were reviewed according to the journal Molecular Ecology:techniques used in molecular ecology,molecular population biology,molecular environmental genetics,molecular adaptation.Experimental results showed that DNA diversity was the richest one in all aspects of genetic diversity.Even in captive populations of the giant panda and natural populations of a precise inbreeder——the wild soybean( Glycine soja )which previously were considered as the species with a low level of genetic variation,experimental results showed that individuals had their uniqueness of genetic composition when sensitive detecting methods were used.Much higher level of DNA diversity within populations and a low level of genetic differentiation among populations indicated that most of polymorphic DNA was caused by neutral or near neutral mutations which was confirmed by the unrelatedness between salt tolerance levels and polymorphic DNAs of individuals in natural populations of wild soybean under saline conditions.A few polymorphic DNAs were found to be related with morphological differentiation, and it showed clinic variation which implied adaptative.Sampling stretegy of monitoring and conserving genetic diversity should be modified to meet the needs of conservation of adaptative characters.Molecular genetics has also applied to workout plans for ecosystem and species conservation.Finally from methodology and philosophy of molecular biology some new ideas and concepts were suggested.One is characterization of basic functioning unit of ecosystems for further gene and gene gene interaction studies.

介绍了这个新学科的基本内容,其特征是DNA标记的应用。结合我国最近几年动植物自然种群的分子研究,介绍国际分子生态学各个领域的进展:①分子生态学的技术;②分子种群生物学;③分子环境遗传学;④分子适应。实验结果显示:只要方法灵敏,DNA具有最高水平的多样性。即使是原先认为遗传变异很少的大熊猫和野生大豆,使用灵敏的方法,也能证实生物个体遗传组成的唯一性。种群内DNA的高度多态性,不同景观生态类型种群之间低水平遗传分化,说明自然种群绝大多数多态DNA位点是中性、近中性突变。至今没有发现盐渍条件下植物个体耐盐性水平与多态DNA有相关性,更证实这一点。发现少数多态DNA位点与形态分化有关或呈明显的地理梯度,暗示其适应意义。自然种群这两种生态学功能不同的多态DNA的存在,说明有必要重新讨论遗传多样性研究和保存中的取样策略。分子遗传研究也指导生态系统和物种的保育。文章最后从分子生物学的方法论和已经阐明的生态过程的众多分子信息提出分子生态学的新思路。建议分解生态系统,找出一个或少数物种和环境构成生态系统的基本功能单位,研究所涉及的基因及基因对基因的相互作用。进一步提议首先分析最简单的生态系统里发生的专一过程的分子细节。

文章综述了以16S rRNA序列分析技术为主的分子生物学技术在反刍动物瘤胃微生态研究中的应用现状,主要包括:序列分析技术、DGGE指纹技术、寡核苷酸探针杂交分析法以及定量PCR等分子生物学方法和技术。今后的发展趋势是加强这些方法间及其与传统方法的有机结合,并发展新的方法,促进瘤胃微生态研究的深入开展。

The gastrointestinal(GI) tract of mammals harbors a large number of microbiota comprised primarily of bacteria,which have a profound influence on nutritional,physiological and immunological processes in the host animal.Traditionally,GI bacteria have been studied via cultivation-based techniques.Recently,some molecular ecology techniques based on the 16S rRNA(DNA) gene have been used widely in studies of gastrointestinal bacterial communities.These techniques include: 16S rDNA gene clone libraries,16S rDNA fingerprinting techniques,quantitative PCR,fluorescent in situ hybridization(FISH) and DNA microarray,et al.Here,a critical summary is provided of current molecular approaches for studying the GI microbiota.

哺乳动物胃肠道中栖息着大量的微生物(主要为细菌),它们在营养、生理、免疫等方面对宿主起着有益作用。传统上,胃肠道菌群研究主要依靠培养技术。近来,一些基于16S rRNA(DNA)的分子生物学技术已被广泛应用于胃肠道菌群的研究,这些技术主要有16S rDNA克隆基因文库、16S rDNA指纹技术、定量PCR技术、荧光原位杂交技术及基因芯片技术等。对这些用于胃肠道微生态研究的分子生物学技术作一综述。
