Objectives To study the effects of glutathione (GSH) and vitamin E on toxicity caused by cadmium.Methods Four groups of Syrian hamsters were injected subcutaneously with 0,35,60 and 70 μmol cadmium chloride(CdCl 2) per kilogram of body weight(kg),respectively.Another two groups of the hamsters were pretreated with GSH and vitamin E,respectively and then injected subcutaneously with 70 μmol/kg CdCl 2.Serum activities of lactic dehydrogenase(LDH) and alanine transaminase(ALT),urine activity of LDH,urine protein,cadmium level in the liver and renal cortex were determined for the animals 24 hours after treatment with cadmium.Results Serum activities of LDH and ALT increased with cadmium dose of exposure,and those were significantly higher in the group with 70 μmol/kg CdCl 2 than in the controls.There was no significant difference in urine LDH and urine protein between groups with various cadmium doses and controls.Cadmium levels in the liver and renal cortex increased with cadmium dose of exposure in various groups.GSH in the renal cortex in the cadmium exposed groups were significantly higher than in the controls,but there was no significant difference in liver GSH between various groups and the controls.Serum activities of LDH and ALT in the hamsters pretreated with GSH and vitamin E before cadmium administration were significantly lower than those with cadmium only,and GSH level in the former was also significantly lower than that in the latter.Cadmium in the renal cortex of the hamsters pretreated with vitamin E was also lower than that in those only with cadmium.Conclusions Acute exposure to cadmium mainly caused liver damage in the hamsters.Pretreatment with GSH and vitamin E can antagonize the liver damage caused by cadmium,which could be attributed to,as postulated,their effects on anti oxidation and stabilization of biological membrane,rather than the changes in GSH level.

目的 探讨GSH和维生素E对镉毒性的影响。方法 给 4组Syrian仓鼠皮下注射 0、 35、 6 0和 70 μmol/kg体质量CdCl2 ,另 2组仓鼠用GSH和维生素E预处理后再投与 70 μmol/kgCdCl2 ,染镉后 2 4小时观察血清LDH、ALT ,尿LDH和尿蛋白 ,肝脏和肾皮质镉含量以及肝脏和肾皮质GSH含量的变化。结果 随着染镉剂量的增加 ,血清LDH和ALT活性有升高趋势 ,70 μmol/kg组仓鼠血清LDH和ALT活性明显高于对照组 ;尿LDH活性和尿蛋白含量各染镉组与对照组之间无明显差异 ;各染镉组仓鼠肝和肾皮质镉浓度随染镉剂量增加而升高 ,肾皮质GSH含量均明显高于对照组 ,而肝脏GSH含量与对照组之间无明显差异。给仓鼠GSH和维生素E预处理后仓鼠血清LDH和ALT活性明显低于单纯染镉组 ;肝和肾皮质GSH含量低于单纯染镉组 ;维生素E预处理组仓鼠肾皮质镉浓度也低于单纯染镉组。结论 急性染镉主要引起仓鼠肝脏损害 ,GSH和维生素E预处理可以拮抗镉的肝脏毒性 ,此种保护作用不是由于GSH含量的改变 ,推测可能与它们的抗氧化和稳定生物膜的特性有关。

It was similar to the normal control group about the numbers of hepatocyte′ apoptosis of the co-treatment group with cadmium and Vitamin E(P>0.01),but it was more less than the cadmium group(P<0.05).2.The apoptotic cells of the hepatocyte had the following characters: (1)The volumes of the cells became smaller. (2)The nucleoli disappeared and the chromatins distributed along the nuclear envelopes.(3)The nuclei appeared pyknosis and karyorrhexis. The analysis of morphometry suggest that the nuclear important parameters of apoptotic cells decreased obviously(P<0.01). The apoptotic bodies were seen in some cells.Conclusion Vitamin E could repress the hepatocyte′s apoptosis with chronically cadmium-exposed.

Objective To explore the protective effect of mice exposed cadmium chronically. Methods Sixty three month old Kunming male mice were randomly divided into three group:(1)Cadmium group(treatment with cadmium);(2)Vitamin E group(treatment with cadmium and vitamin E);(3)Normal control group(treatment with N.S). The morphological change of the spermatogenic cells were observed under the light and electron microscope. The morphometric parameters of the ultrastructures of spermatogonia and spermatozoa were study by stereological analysis. Results (1)Cadmium group:The testes of all the mice were damaged. The injuried degree of the testes may be divided into 3 types:slight,middle and serious degree of injury. Compared to normal control group,the morphometric parameters(e.g.mean profile erea,mean volumn,mean surface area etc)of spermatogonia and suermatozoa nuclei were decreased markedly(P< 0.05). (2)VE group:The testes fine structure of great majority mice were normal,only a few testes had slight and middle degree of injury. Compared to normal control group,the morphometric parameters of spermatogonia and spermatozoa nuclei were near and no significant difference(P >0.05). Conclusion The protective effect of vitamin E on spermatogenic cells injury induced cadmium were prominent.

目的探讨维生素E(VitaminE,VE)对慢性镉中毒小鼠睾丸生精细胞损伤的保护作用。方法3月龄昆明种雄性小鼠60只,随机分3组:镉组(每次CdCl22mg/kg,皮下注射,每周2次,共3个月),VE组(染镉同时给VEl0mg·kg-1·d-1,灌胃)和正常对照组(注射等量生理盐水),用光镜、电镜观察生精细胞形态变化,并对精原细胞和精子的超微结构进行立体定量分析。结果(1)与正常对照组比较,镉组小鼠睾丸均受损,根据受损程度可分为轻、中、重3型;精原细胞胞核和精子头内细胞核的平均截面积、平均体积和平均表面积等形态参数值均显著缩小(P<0.05);(2)VE组:大部分小鼠睾丸的形态结构接近正常,仅少部分小鼠的睾丸受到轻、中型损伤;精原细胞胞核和精子头内细胞核的平均截面积、平均体积和平均表面积等形态参数值与正常对照组的相应值接近,无显著性差异(P>0.05)。结论VE对镉引起的小鼠生精细胞形态的损伤有一定保护作用。
