Investigating the characteristics of plasma proteins adsorbed on polymeric surfaces are of significance in the study of blood compatibility of polymers. ln estimating the amount of adsorbed blood proteins, Fourier transofrm IR internal reflection, isotopic labeling technique and ultraviolet spectroscopy have been used as suggested by Matsui, Lee, and Bull respectively.Generally, for the conformational changes only microcalorimetry and circular dichroism have been used successfully. Based upon the report of 赤池敏宏, we have tried to estimate both of the amount of the adsorbed bovine serum albumin and its conformational changes with ultraviolet spectroscopy.Polypeptide chains have strong absorption peaks at 190 nm. When peptide chains are in α- helix or β-fold, peptide chains are regularly packed according to certain rules.All the orderly peptide chains in which eletrons transited may interfere with each other, so that there is difference of the ultraviolet spectra among the α-helical, β-folded and amorphous states of peptides. The percentage of α-helix can be determined by the difference between the max at 220 nm and the rain at 205 nm. Equations for the calculation of the amount of adsorbed BSA and percentage of conformatioal changes are summaried as follows: BSA adsorbed C (μg/cm~2)=1.29X10~2X(Dmeasured at 202/2) Conformation ε205=1.155X10~5X(Dmeasured at 205/2XC(μg/cm~2))xH=(3600-ε250)/2600 where xH is the content of α-helix.According to above equations, the amount of adsorbed BSA on potyether polyurethane and quartz plate was 0.342 g/cm~2 and 0.322 g/cm~2, and the percentage of a-helix 40% and 44% respectively. The results showed that there was no change of conformation in BSA adsorbcd on polyether polyurethane, which was similar to the findings of 赤池敏宏。As the experiments were done with microanalysis, even traces of impurities would be expected to influence the accuracy of results markedly.

The nature of a polymer surface governs the processes involved in interaction between blood and material. In this paper, we summarize the techniques used to measure surface propertied, review the hypotheses for the blood compatibility of biomaterials, and discuss the unique properties of some common biomedical polymeric surfaces.

本文首先概述了医用高分子表面的表征方法。接着着力讨论材料和血液的相互作用,其中主要阐述目前血液相容性研究所涉及的内容和各种抗凝假设。最后具体介绍了几种常见医用高分子材料的表面特性。
