The experiment for selecting medium was designed with orthogonal method. The cell lines tolerated to 0.5, 0.7,1.0% NaCl were established separately. The regenerated plants from various NaCl tolerant cell lines were obtained. The results are as follows.1.MS + 0.5 ppm 2,4-D+l.0 ppm Zt was the best medium for inducing callus,MS + 0.2ppm IBA+1.0ppm Zt was the best for differentiation and MS + 0.5ppm IBA was the best for rooting.2.Growing of the callus was inhibited by NaCl.The growth of callus decreased significantly in 0.3% NaCl, and decreased very significantly in 0.5% NaCl.Therefore, 0.5% NaCl concentration can be considered as the critical value for salt-tolerance selection.3.γ-rays over 5kR not only caused increase of the cell size but also inhibited growth of the callus.So, 5kR γ-rays can be considered as optimum mutation dose.

取中华猕猴桃(Actinidia chinensis)101品系试管苗叶片为外植体,诱导愈伤组织,将愈伤组织移入含NaCl的MS培养基选择8次,培养基内NaCl的浓度逐步增加,γ射线作为诱变剂,已筛选出耐0.5%、0.7%、1.0%NaCl的变异细胞系,并均再生出完整植株。试验结果表明:1.MS+0.5ppm 2,4-D+1.0ppm Zt是最佳的脱分化培养基;MS+1.0ppm Zt+0.2ppm IBA是最佳的分化培养基;生根培养基可采用MS+0.5ppm IBA。2.0.5%NaCl可作为耐盐筛选的盐分临界值。3.γ射线处理对愈伤组织的生长及细胞的大小均有显著的影响,5kR可作为变异体筛选的诱变照射量。

In this paper, 5 wheat salt tolerant mutants(H8706-34、H8706-44、H8706-48、RH8706-49、H8706-57)derived from anther culture、EMS induction and salt tolerance selection and their parents (Punong3665、Bainong3039)were used as materials, all the mutants have inherited stably for 9 generations. Differences were revealed between the mutants and their parents using chemical marker(gliadin)and molecular marker(RAPD), the results showed that compared with the parents, the mutants varied not only on the protein level,but also on the DNA level,which supplied hard evidence of the truth of the mutants and ruled out the possibility of salt adaptation. RAPD analysis were conducted among the 5 mutants by 218 primers,which proved they were a series of near isogenic lines of different salt tolerance because of their little difference and similar genetic background.

The result showed that compared with the original line, the mutants varied not only on the salt tolerant indexes level, but also on the DNA level, which provided a strong evidence for the truth of the mutants. This study showed that these mutants (N 1,T 3,T 4,T 5) were salt tolerant new lines of Hordeum brevisubulatium(Trin.)Link.

以经60 Co物理诱变种子、EMS诱变和耐盐反复筛选后已稳定 6代的短芒大麦耐盐碱突变体N1、T3 、T4、T5及其亲本野生短芒大麦为材料 ,进行耐盐碱生理生化指标 (脯氨酸、质膜透性 )测定及RAPD分析。发现突变体与野生型相比 ,不仅在生理生化指标上有明显的差异 ,而且在DNA水平上发生了突变。研究结果为耐盐碱突变体的真实性提供了有力的证据 ,表明突变体N1、T3 、T4、T5是耐盐碱短芒大麦新品系。
