本文介绍用细胞-载体共交联法制备固定化葡萄糖异构酶的方法。有机及无机类载体具有良好的交联特性。将其与含葡萄糖异构酶的游动放线菌菌体混合,在一定条件下用戊二醛交联,可制备具有良好性能的固定化酶。应用正交试验法,选出了适合于该载体的交联条件。因此细胞-载体共交联法制备的固定化酶,得率比无载体法提高三倍,生产成本降低约60%。该固定化酶的柱式连续转化最高活性为6700u/g。异构化生产能力为6.6公斤/每克绝干固定化酶。对该固定化酶的一般酶学性质及工作性能在本文中也做了介绍。

Numerous enzymatic, electrochemical, immobilized chloroplasts and immobilized microbial cells methods have been reported, of the methods, enzymatic method has been usedin the industrial production as the principle of this method is simple and the reactor costs is not expensive. ImcnobiLized microbial cells is also used to regenerate, but the selectivity of the reaction is less than with enzymatic methods and their reactor costs more than the former. Although electrochemical and immobilized chloroplast methods are possible to use for regeneration of NAD(P)H cofactor, they have not been reported in the practical application as yet.

固定化酶的制备及其性质己在世界范围内引起人们的极大兴趣,而这种兴趣主要的体现在它们的工业生产方面。NAD(P)H再生是进行酶促合成及氧化还原反应的重要限制性因素之一。已有大量利用酶学的、电化学的、固定化叶绿体及固定化微特征细胞方法进行NAD(P)H再生的报道。在这些方法中,酶学方法由于它们原理简单、反应器造价也不太昂贵己被应用于工业生产之中;固定化微生物细胞虽然也被用于工业生产,但它们反应的选择性较差且反应器的造价也较前者为高;虽然电解法和固定化叶绿体有可能用于NAD(P)H的再生,但直到目前为至仍未见到有关实际应用的报导。
