A procedure of shoot-apex grafting in vitro of mulberry is described. Anaverage frequency of 75%~80%of successful shoot-apex grafting of several major com-mercial varieties was achieved by using 2 cm segments from the seedlings as rootstocksand l~2mm shoot apices from adult plants as scions,It is concluded that thickening thecircumference of rootstock stems is the most critical factor for successful shoot-apexgrafting in vilro of mulberry.

在无菌条件下将桑籽接种至含MS无机盐与0.2%凝胶剂(Gelrite)的培养基上,于26℃每日16h、10001x照明下培养;2周后以激素处理茎切段,转培于蛭石培养基,10~12d苗茎围度达8mm时,以长2cm的切段为砧木,以707等桑品种的1~2mm茎尖作接穗,嫁接成活率达75%~80%。增大砧木苗基的围度是取得桑树茎尖嫁接成功的关键。

我国柑桔生物技术研究已有20几年的发展,主要涉及4个方面:①胚和胚乳培养以及茎尖微芽嫁接等以解决有种和生产问题。②系统建立的基础性工作:原生质体培养与生理研究。③人工创造种质的研究:原生质体融合与转基因。④分子标记技术在柑桔研究中的应用。本文继《我国柑桔离体培养研究的进展》(1989)一文的内容之后,对近10多年来我国柑桔生物技术的进展作了全面的介绍。
