Tong-mai-ling has been used by us with success clinically in the treatment of superficial corneal scars since 1975. Studies commenced after corneal scars were successfully produced in both eyes of 10 rabbits. Tong-mai-ling was instilled to the left eye of each rabbit. 3 times a day. and injected subconjunctivally once a week, for 3 months, with the right eye serving as control. At the end of 2 weeks after completion of treatment, scars of the treated eyes were found to be diminished both in size and thickness, and the stromal cells became smaller and more slender, arranging in a more orderly fashion. The thickness of the cornea became normal or nearly normal too. and its acid polymu-cosaccharide content declined to normal as well. Scars of the non-treated right cornea showed no change, and the stromal cells were found to be increasing in number and size and irregularly arranged. The thickness of scar and neutral polymucosaccharide content of the untreated corneas were also found to be increased.The above observations seemed to illustrate that Tong-mai-ling has its action mainly on the stromal cells of the corneal scar tissue. The drug appeared to have a certain inhibitory effect on the proliferation of the collagen-forming cells and synthesis of collagen fibers, and induced the recovery of the lowered acid polymucosaccharide content around the collagen fibers.

以家兔制作角膜瘢痕模型后,左眼用通脉炅治疗,3个月后,角膜瘢痕缩小、变薄,角膜基质细胞体积小而细长,排列较规则,角膜厚度正常或接近正常,间质中酸性粘多糖接近正常。作为对照组的右眼瘢痕无明显变化。认为通脉灵治疗角膜瘢痕的有效机理主要是影响角膜基质细胞的数量和功能。药物对胶原形成细胞的增殖及胶原纤维合成有一定抑制作用,并能使减少的胶原纤维周围的酸性粘多糖恢复。

IH764-3, an effective component isolated from Salvae milliorrhize (a component of traditional Chinese medicine) was used against pulmonary fibr-osis in this study. The results indicated that in the treated group, lung coefficient, surfactant, hydroxyproline content and FGF activity were significantly lower than those in the model group. All the results demonstrated that IH764-3 has prophylactic and therapeutic effects on the development of pneumonitis and pulmonary fibrosis.

本文主要介绍丹参提取的有效单体(IH764-3)抗肺纤维化的实验研究。结果表明IH764-3治疗组动物的肺系数,羟脯氨酸,表面活性物质含量,FGF活性均明显低于模型对照组。电镜观察证实治疗组肺胶原形成细胞数量,炎性细胞渗出,胶原纤维和弹力纤维都较模型组明显减少,肺泡毛细血管和肺泡膈内小血管形态较正常,Ⅱ型肺泡上皮细胞的数量和形态较正常。以上结果证明IH764-3对博莱霉素诱致的肺炎、肺纤维化具有明显的预防和治疗作用。

The studies reported here showed that the fibroblasts and stromal cells factually synthesized and secreted Fn and TSP which were found to appear on the cell surface and interspace among the adjacent cells as well as the extracellular fibrous network matrix,demonstrating some bulky patch patterns.In addition,both the fibroblasts of rabbit corneal scars and the stromal cells of mouse bone marrow in long-term culture contain fibronectin and thrombospondin in their cytoplasms. Application of immunoelectron microscopic technique makes it possible to study the localization and distri- bution of Fn and TSP in the varied diseases concerning the connective tissue hyperplasia.It could provide a direct means to present visual morphological evidence about the mechanism of fibrosis formation and the interaction of collagen-forming cells,stromal cells,and other type cells.

本文用免疫电镜技术(过氧化酶标记和胶体金标记)在来自兔角膜瘢痕的纤维母细胞和来自小鼠骨髓的长期培养的基质上进行了纤维粘连蛋白(FN),血小板凝血酶敏感蛋白(TSP)定位。结果显示在纤维母细胞和骨髓基质细胞表面以及细胞外细纤维构成的网中。另外,有时在上述二种细胞胞浆中也可以看到FN和TSP的定位。应用免疫电镜技术可以研究各种结缔组织增生性疾病,FN和TSP的分布以及它们和结缔组织增生的关系,为纤维化形成机制以及胶原形成细胞、基质细胞和其它细胞的关系提供了形态学依据。
