On the 6th day in vitro, the culture medium was substituted by Earle's ballanced salt solution without glucose, then the cells were incubated in an atmosphere of 95% nitrogen and 5% CO_2. After 1 to 4 hours, increasing number of nerve cells became swollen, lost their character of optical refraction under phase contrast microscope, and were stained with trypan blue. Their leakage of LDH increased and their ability of high affinity uptake of GABA decreased. The reduction of GABA uptake took place earlier and more remarkable than that of other parameters. No significant change was observed if the ceils were incubated with glucose but without oxygen or with oxygen but without glucose. The above mentioned alterations were similar to nerve cell changes following cerebral ischemia in vivo. This model in vitro is useful in the study of ischemic nerve cell damage, their recovery and the protective effect of drugs.

Results:NGF could increase the number of nerve cells that had been cultured for 4d in vitro;aFGF promoted the processes and neurite growth of neurons in the same culture state.Both NGF and aFGF could enhance the development of cell bodies. When NGF and aFGF were both used in the medium,the growth of cultured neurons was better than when only one of them was used. Conclusion:NGF and aFGF have specific and coordinated actions on the growth and the development of cultured spinal cord neurons,and conbinated usage two kind trophic factors can produce better results.

The toxic effects of glutamic acid on nerve cells and the treating effects of Congsheng Capsules (CC) were investigated. The method of rat cerebral cell culture was used in this experiment. Changes in the levels of LDH and MDA in the nerve cell culture fluid, and the level of Ca 2+ in the nerve cells were observed after glutamic acid and CC were used to the nerve cells respectively. The results showed that glutamic acid had toxic effects on the nerve cells; and CC could reduce the leakage of LDH and the excessive production of MDA, and decrease the level of Ca 2+ . The results suggest that CC have certain protective effects on the nerve cells.
