Objective To demonstrate the role of vitamin K 2 on proliferation and differentiation of osteoblasts. Methods Ostoblasts from newborn rat calvaria were cultured for 3 days in a medium containing various concentrations of vitamin K 2. The cell count, H 3 thymidin incorporation and intra cellular osteocalcin (bone γ carboxyglutamic acid containing protein, BGP) level were studied to observe the effect of vitamin K 2 on osteoblasts. Results In the experiment of adding vitamin K 2 10 -5 ,10 -6 ,10 -7 and 10 -8 mol/L, the osteoblast count was 28×10 5 , 26×10 5 , 29×10 5 and 30×10 5 /ml,respectively. BGP was 38 12±6 47, 46 65±4 59, 34 38±5 22 and 32 22±4 86ng/10 6 cell respectively. While in the control group they were 32×10 5 /ml and 19 92±3 67ng/10 6 cell. The results showed that the proliferation of osteoblast was slightly suppressed and the cellular BGP level was evidently increased by vitamin K 2. Conclusions Vitamin K 2 may be used as a new medicine for treatment of osteoporosis.

Results\ The levels of BGP, ALP, IL 6 in post menopausal women were significantly higher than those in the control group ( P <0 01), the values of U HOP/Cr and U Ca/Cr were significantly higher than those in the control group( P <0 01), whereas the BMD and the level of E 2 in postmenopausal women were significantly lower than those in the controls( P <0 01). There was significant differences in BMD, E 2, IL 6, BGP,ALP,U HOP/Cr and U Ca/Cr between OP and NOP groups. Conclusion\ There is close relationship between osteoporosis and E 2 and IL 6. Decreased expression of E 2 and increased expression of IL 6, is a possible important reason for acceleration of bone resorption.

目的 了解E2 和IL 6在骨质疏松症发病中的作用。方法 选择绝经后妇女 12 0例 ,绝经后有骨质疏松者 6 0例 (OP组 ) ,绝经后无骨质疏松者 6 0例 (NOP组 ) ,另外选择绝经前妇女 6 0例为对照组。对 180名妇女雌二醇 (E2 )、骨密度 (BMD)、白细胞介素 6 (IL 6 )、血清总碱性磷酸酶(ALP)、骨钙素 (BGP)、尿羟脯氨酸肌酐比值 (尿Hoc/Cr)、尿钙肌酐比值 (尿Ca Cr)等指标进行了测定。结果 绝经后妇女骨形成指标BGP及ALP明显高于对照组妇女 ,其中ALP在OP组和NOP组间有差异 ,而BGP在OP组和NOP组间无差异 ;绝经后妇女骨吸收指标尿HOP/Cr及尿Ca/Cr明显高于对照组妇女 ,OP组尿HOP/Cr及尿Ca/Cr又明显高于NOP组 ;绝经后妇女的血清E2 的含量明显低于对照组 (绝经前妇女 ) ,OP组又明显低于NOP组 ;绝经后妇女血清IL 6的含量明显高于对照组妇女 ,而OP组又明显高于NOP组。结论 本研究证明E2 、IL 6与骨质疏松关系密切 ,雌激素水平的下降 ,IL 6分泌增多 ,是导致骨吸收加速的重要原因之一

Objective To study the effect of 2β-(3-hydroxypropoxy)-Calcitriol (ED-71) on the cell growth and differentiation of human osteoblast-like cell line OS-732. Methods Osteoblast-like cells were cultured with various concentration of ED-71.The effects of ED-71 on proliferation and differentiation were detected by (MTT), alkaline phosphatase activity and level of osteocalcin (bone gla protein,BGP). Results The proliferation rate of osteoblast-like cells cultured for 4 days was significantly inhibited by 10~ -7mol/L concentration of ED-71. ED-71 in 10~ -8, 10~ -9mol/L concentration can significantly increase BGP level, but ED-71 in 10~ -7mol/L concentration may decrease BGP level.Alkaline phosphatase activity compared with control was not changed significantly. Conclusions ED-71 can inhibit cell growth in high concentration and improve cell differentiation in lower concentration.

目的观察2β(3羟丙氧)骨化三醇(ED71)对体外培养人成骨样细胞增殖与分化的影响,进一步探讨ED71治疗骨质疏松的机制。方法采用塞唑蓝(MTT)分析法检测ED71对体外培养OS732人成骨样细胞增殖作用与效价。采用培养细胞匀浆测单位蛋白碱性磷酸酶活性,培养基中骨钙素含量分析及原位杂交方法检测ED71对OS732人成骨样细胞分化的影响。结果培养第4天,10-7mol/LED71显著抑制成骨样细胞增殖(P<0.05),而10-8mol/L、10-9mol/LED71对成骨样细胞的作用不明显。培养4d后培养基中骨钙素水平在10-7mol/LED71组变化不大,在10-8mol/L、10-9mol/LED71组骨钙素水平显著增高(P<0.05或P<0.01)。原位杂交转化生长因子β(TGFβ1)表达明显增加,而碱性磷酸酶比活性无明显变化。结论ED71抑制成骨样细胞增殖高剂量作用显著,促进成骨样细胞分化较低剂量显著。
