Objective To investigate the effect of recombinant human bone morphogenetic protein-2 sustained release hydrogel microspheres (reBMP2-dex-HM) on the proliferation and differentiation of osteoblasts.Methods The secondary culture osteoblasts were divided into three groups according to the different ingredients being added into the DMEM culture medium:rhBMP-2 group (A),dex-HM group (B) and rhBMP2-dex-HM group (C).The proliferation of the cultured osteoblasts was measured by using cell counting method,MTT method and flow cytometry.The contents of bone BGP secreted by osteoblasts were also measured by RIA method.Results The in vitro cellular study showed no significant difference in the cell number and cell viability on the second day after plate culture,but 2 days later the cell number and cell viability in group A were significantly increased as compared with those in other groups (P<0.05).Six to 8 days after plate culture,the cell number and cell viability in group C were increased as compared with other two groups (P<0.01).After 14 days,the cell number in groups A,B and C was ( 22.97± 0.23),( 13.89± 0.57) and ( 32.46± 0.67)×104 cells/ml,with the difference being significant among them (P<0.01).The flow cytometry revealed that the G_2/M+S percentage in group C were the highest 4-8 days after plate culture (P< 0.05).Among the three groups,the content of BGP in group C was the highest.Conclusion The rhBMP2 -dex-HM can promote the proliferation and differentiation of osteoblasts through a long period of controlled release of rhBMP-2.Hydrogel microspheres may be an ideal controlled release carrier for growth factors.

Objective To study the forming condition of water-in-oil(W/O) salbutamol sulfate nanoemulsion and investigate its potential as a peroral administration carrier.Methods Effect of preparation technology on nanoemulsion formation was investigated.Using pseudo-ternary phase diagrams,effect of soybean oil and Span 80 ratio on the formation field of nanoemulsion was studied.A HPLC method was established to analyze the content of salbutamol sulfate in the nanoemulsion by solvent extraction.Absorption of the nanoemulsion was investigated using an in-situ intestine circulation method in rats.Results The preparation technology was found to influence the formation of nanoemulsion significantly.A stable and practical water-in-oil nanoemulsion formulation with a higher water content was obtained,that is,soybean oil∶Span 80∶Tween 80∶water 22.8∶22.8∶45.5∶8.9(w∶w).In addition,salbutamol sulfate was incorporated into the aqueous phase and then water-in-oil(W/O) salbutamol sulfate nanoemulsion was prepared.The in-situ absorption experiment in rats demonstrated that salbutamol sulfate was released from the nanoemulsion in a controlled manner.Conclusions Water-in-oil nanoemulsion is a potential controlled release carrier for water-soluble drugs.

目的研究硫酸沙丁胺醇油包水(W/O)型口服纳米乳的成乳条件及小肠吸收情况。方法采用两种制备方法考察不同制备条件对纳米乳形成的影响;采用伪三元相图法考察油相与司盘80的不同配比对成乳区域的影响,求出W/O型纳米乳形成区域;采用HPLC法测定纳米乳中硫酸沙丁胺醇的含量;采用大鼠在体小肠循环法初步考察该纳米乳的口服吸收情况。结果在本实验范围内,稳定且含水量较高、有实际应用价值的W/O型纳米乳的优化处方为豆油、司盘80、吐温80、蒸馏水的质量比为22.8∶22.8∶45.5∶8.9。根据最优处方,将水溶性药物硫酸沙丁胺醇溶于水相中,制得了W/O型纳米乳。大鼠在体小肠循环吸收试验初步证明了该纳米乳具有缓释作用。结论W/O型纳米乳可作为水溶性药物的缓控释制剂的载体。
