We have reported previously that actinomycin K possessed marked inhibitory action against Ehrlich ascites carcinoma and 3 other types of mouse sarcomas. We have reported previously that actinomycin K possessed marked inhibitory action against Ehrlich ascites carcinoma and 3 other types of mouse sarcomas. In the present paper its effect on the mitosis of Ehrlich ascites carcinoma cells was studied. 28 mice inoculated with Ehrlich ascites carcinoma cells were divided into 3 groups. Two of the groups were injected intraperitoncally with a single dose of actinomycin K 50γ and 200γ/kg respectively, and the third group was injected with normal saline which served as control. At the time before and 3, 6, 12 and 24 hours after injection, the percentages of mitotic and of degenerative cells, and the concentration of carcinoma cells were examined. It was found that after administration of actinomycin K there were significant changes. The percentage of degenerative cells was increased. The cell concentration and the percentages of all 4 stages of mitosis were significantly decreased, the metaphase being affected most strikingly. The maximum effect was observed at six hours after injection, and a return to the normal occurred in 12 hours. These changes were more marked in the group given 200γ/kg than that given 50γ/kg. When actinomycin K was injected intraperitoneally at the dosage of 50γ/kg/day for 7 days in mice bearing Ehrlich ascites carcinoma, the cytological changes were similar to those given single injections, but the effects were more significant.

(一)放綫菌素K以50及200微克/公斤对Ehrlich腹水癌小白鼠一次腹腔注射,于給药前及給药后3、6、12、24小时的不同間隔时間,抽取腹水进行检查,結果发現,于給药3小时后,对癌細胞的分裂象和腹水癌細胞浓度,均可明显地減低,退变細胞的百分比增高,以在6小时为最,12小时后趋向恢复。进行分裂象分期計数时,对各期都有显著的影响,以对中期分裂象抑制最为明显。 (二)放綫菌素K50微克/公斤,每天注射一次,連續7天,也能使癌細胞浓度和各期分裂象明显減少,其作用較一次給药組更为显著。

In this paper the antitumour action of 17 phosphoramide nitrogen mustard compounds were studied. It was found that bis-(1-aziridinyl)-[bis-(2-chloroethyl)-amino]-phosphine oxide (AT-222) possessed a marked antineoplastic activity.The main results were as follows: 1.Intraperitoneal injections of 1—2.5 mg/kg of AT-222 produced a marked inhibitory action on mouse sarcoma 180, reticulo-cell sarcoma (L-2), sarcoma AK, spindle cell sarcoma and rat Jensen sarcoma and Walker carcinosarcoma....

Intragastric administration of this drug brought about a definite inhibition on the growth of Jensen sarcoma in rats. No effect on Ehrlich carcinoma (both ascitic and solid forms) was observed when the drug was injected intraperitoneally. 2.In mice, the acute and subacute LD_(50) of AT-222 were found to be 19 and 4.7mg/kg respectively. In rats the subacute LD_(50) was 2.7mg/kg (observation period= 10 days) or 1.9mg/kg (observation period=17 days) 3.In dogs, intravenous injections of AT-222 at 0.1 mg/kg/day for 10 days caused no significant change in blood picture, urinalysis, stool routine examination and body weight. In the group given 0.3 mg/kg, white cell counts were slightly decreased on the 5th day after drug administration, and gradually recovered after cessation of the drug. Pathologic studies revealed hemorrhagic spots in the intestinal mucosa and pyknotic nuclei in the granulosa cells and in the lymphocytes of the spleen.In the group of 0.9 mg/kg, white cell counts were markedly decreased and the animals died 12--14 days after drug administration, In pathologic examinations diffuse hemorrhage in the internal organs and atrophy of gastric and intestinal glands, adrenal bodies and testis were seen.

本文研究了17种磷酰胺氮芥类化合物,发現N-双(2-氯乙基)-N'-N"-二乙撐基磷酰胺(以下均簡称AT-222)具有較强的抗癌作用。主要結果如下:(1)腹腔注射AT-2221—2.5毫克/公斤对小白鼠肉瘤180,网織細胞肉瘤,AK肉瘤,梭形細胞肉瘤,大白鼠Jensen肉瘤和Walker癌肉瘤都有明显的抑制作用,口服給药时对Jensen肉瘤也有一定的疗效。但对Ehrlish癌固体型和腹水型則无明显的影响。(2)AT-222对小白鼠的急性和亚急性LD_(50)分別为19和4.7毫克/公斤,大白鼠的亚急性LD_(50)为2.7毫克/公斤(停药后观察3天)和1.9毫克/公斤(停药后观察10天)。(3)給狗靜脉注射AT-222 0.1毫克/公斤,其血象、大小便和体重均无明显改变。在0.3毫克/公斤組,給药第5天时白血球稍有下降,停药后逐渐恢复,解剖发現腸粘膜有出血点,脾淋巴細胞核和卵巢顆粒細胞核有固縮現象。0.9毫克/公斤組,于給药后第5天时白血球卽明显下降,停药后2—4天动物相继死亡,解剖检查发现各脏器均有弥蔓性出血,腸、胃腺体、腎上腺和睾丸均有明显萎縮。

The lethal effect of deep freezing on the tumor cells was investigated on the mouse Sarcoma 180 (S—180) and mouse Ehrlich ascite (ECA) cells.The homogenates of S-180 or ECA ascite were frozen at various temperature ranging between 0~-196℃ for 3 minutes and thawed thereafter.In another experiments, the freezing—thawing process was repeated 3 times successively.The treated tumor cells were then inoculated into normal mice to evaluate the viability of the cells.The results of the experiments...

The lethal effect of deep freezing on the tumor cells was investigated on the mouse Sarcoma 180 (S—180) and mouse Ehrlich ascite (ECA) cells.The homogenates of S-180 or ECA ascite were frozen at various temperature ranging between 0~-196℃ for 3 minutes and thawed thereafter.In another experiments, the freezing—thawing process was repeated 3 times successively.The treated tumor cells were then inoculated into normal mice to evaluate the viability of the cells.The results of the experiments indicated that both S-180 and ECA cells did not lose their viability after a single freezing treatment in temperature range 0~-196℃ as evidenced by the growing tumors in the inoculated mice.After 3 repeated freezing,the S-180 cells lost their viability completely on freezing temperature -40~-196℃,but for ECA cells,the lethal temperature descended to -140~,-196℃.Therefore,it seems that the ability of the endurance for the freezing deterioration is different in various tumor types.The lethal effect of 3 repeated treezing treatment is stronger than that of a single one.

本文研究了不同温度的冷冻处理对小鼠肿瘤细胞的杀伤效应.小鼠 S180肉瘤匀浆细胞经过不同低温冷冻三分钟后立即升温,并接种到正常小鼠中,结果0—-196℃的各组冷冻处理均不能达到完全杀伤肿瘤细胞的效果.如连续冻融三次,则在经过-40—-196℃处理的各组细胞接种后均未在小鼠中产生肿瘤.同样,小鼠艾氏腹水癌的腹水,经过0—-196℃三分钟的冷冻处理,不能完全杀伤其肿瘤细胞;但经三次冻融,并降温到-140℃或更低,才能使其细胞完全失去活力.因此,不同种类的肿瘤细胞对冷冻的忍受能力是不同的.冻融三次要比一次对肿瘤细胞有更强的杀伤作用.
